Self-Assembly Enhances the Strength of Fibers Made from Vimentin Intermediate Filament Proteins

Abstract: Hagfish slime threads were recently established as a promising biomimetic model for efforts to produce ecofriendly alternatives to petroleum polymers. Initial attempts to make fibers from solubilized slime thread proteins fell short of achieving the outstanding mechanics of native slime threads. Here we tested the hypothesis that the high strength and toughness of slime threads arise from the ability of constituent intermediate filaments to undergo a stress-induced α-to-β transition. To do this, we made fibers… Show more

“…Starting with either hagfish protein from the slime glands or the related intermediate filament vimentin, produced by recombinant bacteria, the researchers freeze-dried the proteins and dissolved them in acid, then dripped the solution on top of a buffer. It readily formed a film, and when the researchers picked it up with forceps it came up as thread bundles (4,5).…”

“…Either version, says Rainey, might be useful for imparting flexibility or adding strength to materials, such as car parts, without adding weight. Already, the vimentin fiber's properties are comparable to human tendons, leading Pinto and Negishi to suggest that they might help replace damaged ones (5,6).…”

“…Membrane peptides are often organized in bundles, whose structure is dominated by α-helical coiled-coils 20,[27][28][29][30] . Coiled coils consist of α-helices wind together to form a ropelike structure stabilized by hydrophobic interactions, and this structure is found in about 10% of the proteins in the human genome 31 .…”
