Abstract: Paracoccidioidomycosis (PCM) is the most prevalent deep mycosis in Latin America and is caused by fungi from the Paracoccidioides genus. Virulence factors are important fungal characteristics that support the development of disease. Aspartyl proteases (Saps) are virulence factors in many human fungal pathogens that play an important role in the host invasion process. We report here that immunization with recombinant Sap from Paracoccidioides brasiliensis (rPbSap) imparted a protective effect in an experimental… Show more

“…Such variations may be associated with the differential expression of proteins such as secreted aspartyl proteinases (Sap) and hyphal cell wall protein (Hwp) that are important in the colonization of Candida spp. in the oral cavity (Lie Tobouti et al 2016;Castilho et al 2018). Our present study showed that monospecies ALC3 and C. tropicalis formed hyphae when grown in RPMI-1640.…”

“…Moreover, the increasing of conidia susceptibility after interaction with murine macrophage was also observed after treatment of F. pedrosoi with ritonavir, nelfinavir and indinavir 13 . Castilho et al 30 showed that pepstatin A modulated the interaction between Paracoccidioides brasiliensis and THP-1 macrophages. As observed in P. verrucosa assay with HIV-PIs, yeasts of P. brasiliensis treated with pepstatin A were more susceptible to human macrophages than untreated cells.…”

“…P. brasiliensis showed 53 ORFs encoding proteases, including proteasome subunits, aspartyl, cysteine, metallo-, and serine proteases [102]. However, only a few Paracoccidioides proteases have been characterized over the last two decades; these proteases include serine protease and aspartyl proteases (PbSap) [103][104][105][106]. PbSap is a secreted protease that has been reported in culture supernatants [103,104], and its expression increased in the highly virulent isolate Pb18 and decreased after several generations in culture [104].…”

PbSap is a secreted protease that has been reported in culture supernatants [103,104], and its expression increased in the highly virulent isolate Pb18 and decreased after several generations in culture [104]. In addition, a vaccination strategy using a recombinant version of PbSap was able to confer some level of protection against the lung parenchyma lesions inflicted by P. brasiliensis in a PCM experimental model [104].…”
