Secoisolariciresinol Diglucoside Delays the Progression of Aging-Related Diseases and Extends the Lifespan ofCaenorhabditis elegansvia DAF-16 and HSF-1

Abstract: Secoisolariciresinol diglucoside (SDG) is a phytoestrogen and rich in food flaxseed, sunflower seeds, and sesame seeds. Among the beneficial pharmacological activities of SDG on health, many are age related, such as anticancer, antidiabetes, antioxidant, and neuroprotective effects. Thus, we investigated if SDG had an effect on antiaging in Caenorhabditis elegans (C. elegans). Our results showed that SDG could extend the lifespan of C. elegans by up to 22.0%, delay age-related decline of body movement, reduce … Show more

“…The plates were supplemented with 20 μ M of 5-fluoro-2'-deoxyuridine (FUdR) to inhibit oviposition and hatching. The day when Caenorhabditis elegans was transferred to the plates was recorded as the 0th day of the experiment [ 17 ]. To ensure that trigonelline retained its potency throughout the entire experiment, worms were transferred to fresh plates with or without trigonelline every other day.…”

“…For the pathogenic stress test, the pre-treatment was the same as high temperature resistance test. On the 10th day of the adulthood, the worms were transferred to the NGM plates covered with bacteria Pseudomonas aeruginosa [ 17 ]. The death of worms was observed and recorded every 12 hours.…”

“…Lu et al confirmed the promising attributes of the phytoestrogen secoisolariciresinol diglucoside (SDG) with regard to the health of Caenorhabditis elegans. The authors found that SDG can prolong the lifespan of C. elegans by up to 22.0%, decrease the lethality of oxidative stress and heat, reduce body movement decline linked to age, assuage dopamine nervous deterioration encouraged by 6 hydroxydopamine (6-OHDA), and reduce the toxicity of Aβ protein in C. elegans [115]. Resveratrol, a polyphenol in red wine, is a well-researched biomolecule that is reported to improve brain health by acting as an inhibitor of beta amyloid protein aggregation [116,117].…”

“…As lifespan extension is closely associated with enhanced survival under exposure to different stressors [55], next, we examined whether Reb A supplementation altered the capacity of worms to withstand heat stress and oxidative stress. After exposing 5-day-old adult worms to heat stress (i.e., 35 • C for 5 h) and following a recovery period at 20 • C for 96 h, there was no obvious difference in survival rates between Reb A-treated group and control group ( Figure 3A).…”
