SDS-PAGE analysis of bud-forming cotyledons of Pinus ponderosa

Abstract: The placement of Pinus ponderosa embryos on a benzyladenine (BA) supplemented medium induces the differentiation of two distinct morphological types of cotyledons. Cotyledons in contact with the medium (lower cotyledons) do not elongate, and rapidly respond to BA by forming multiple buds along their entire length. Cotyledons not in contact with the medium (upper cotyledons) elongate, do not form buds, and are similar both morphologically and in protein profiles to seedling cotyledons that do not respond to BA.… Show more

“…2). The overall pattern of variations revealed after seven days of culture by RDD analysis is in good agreement with previous results obtained with less resolving, protein-based approaches [7,[12][13][14]19], and indicates that hormoneindependent transcriptional modulation (mainly downregulation) is the most common response. The predominating event during early phases of in vitro culture thus appears to be a reprogramming of gene expression that occurs regardless of the particular developmental fate of the explants.…”

“…In fact, the largest fraction of differential bands (58%) was found to correspond to mRNAs that are expressed in to control cotyledons and equally disappear after the establishment of either shoot or callus programmes. This hormone-independent downregulation most likely reflects the de-differentiation process which accompanies the initial stages of in vitro culture and involves, among others, a striking alteration of cotyledon physiology with the concomitant negative modulation of several chloroplast proteins [7,12,19,40]. Within the same mRNA population, a smaller fraction of amplicons corresponding to newly expressed genes (28%) was detected.…”
