MTA1 drives malignant progression and bone metastasis in prostate cancer

Abstract: Prostate cancer often metastasizes to the bone, leading to morbidity and mortality. While metastasis‐associated protein 1 (MTA1) is highly overexpressed in metastatic tumors and bone metastatic lesions, its exact role in the development of metastasis is unknown. Here, we report the role of MTA1 in prostate cancer progression and bone metastasis in vitro and in vivo. We found that MTA1 silencing diminished formation of bone metastases and impaired tumor growth in intracardiac and subcutaneous prostate cancer xe… Show more

“…We investigated the prognosis significance of the upregulated and druggable genes. Excluding those genes with no clinical relevance, and those which have already been associated with PCa, such as Ctsb, Gpx2, Idh1, and Nos2 [60][61][62][63][64][65][66], we selected the antioxidant enzyme Srxn1 that had a strong correlation with patient outcome and has no previous related functional studies in PCa.…”

“…Epidermal growth factor cytoplasmic domain (EGFcyt) was identi ed as an inducer of lysosomal cathepsin B expression in thyroid and glioma carcinoma cells [54]. Metastasis-associated protein (MTA1) inhibits cathepsin B expression in prostate cancer [55]. By mRNA sequencing and functional validation, CD147 was identi ed as a novel regulator of cathepsin B, which mediated CD147-faciliated collective invasion in HCC.…”

“…Prostate cancer cells, DU145 and PC3M, were maintained in RPMI-1640 media containing 10% FBS in an incubator at 37 °C with 5% CO 2 as described previously [ 3 , 6 , 8 , 9 , 10 , 11 , 12 , 13 , 14 , 15 , 16 , 33 , 34 , 36 ]. Cells were authenticated using short tandem repeat profiling at Research Technology Support Facility, Michigan State University.…”
