IGF2‐derived miR‐483‐3p associated with Hirschsprung's disease by targeting FHL1

Abstract: HSCR (Hirschsprung's disease) is a serious congenital defect, and the aetiology of it remains unclear. Many studies have highlighted the significant roles of intronic miRNAs and their host genes in various disease, few was mentioned in HSCR although. In this study, miR‐483‐3p along with its host gene IGF2 (Insulin‐like growth factor 2) was found down‐regulated in 60 HSCR aganglionic colon tissues compared with 60 normal controls. FHL1 (Four and a half LIM domains 1) was determined as a target gene of miR‐483‐3… Show more

“…With the development of high-throughput sequencing technology and in-depth research, more and more ncRNAs, especially miRNA and lncRNA, have been found to play very important roles in epigenetic regulation and take part in the occurrence and development of multiple diseases (Moradimotlagh et al, 2019;Pan et al, 2020;Tai et al, 2020). Although there have been some reports about ncRNAs in HSCR, its mode of action and mechanism still need further study (Gunadi et al, 2019;Zhi et al, 2018). In this study, our research team detected that in the stenosis tract of HSCR patients, RMST and miR-1251 were down-regulated apparently.…”

“…SH-SY5Y cells were cultured under 37°C and 5% CO 2 in DMEM medium supplemented with 10% FBS, 100 U/mL penicillin, and 100 μg/mL streptomycin [ 10 ]. Cell models were constructed by the addition of inhibitors and agonists: a Rac1, Limk1, and Cofilin inhibitor group; a Rac1 overexpression group (the PDGF-BB group); a Rac1 overexpression group + a Limk1 inhibitor group (the P-B group); and a Rac1 overexpression group + a Cofilin inhibitor group (the P-C group).…”
