GDF15 is a heart‐derived hormone that regulates body growth

Abstract: The endocrine system is crucial for maintaining whole‐body homeostasis. Little is known regarding endocrine hormones secreted by the heart other than atrial/brain natriuretic peptides discovered over 30 years ago. Here, we identify growth differentiation factor 15 (GDF15) as a heart‐derived hormone that regulates body growth. We show that pediatric heart disease induces GDF15 synthesis and secretion by cardiomyocytes. Circulating GDF15 in turn acts on the liver to inhibit growth hormone (GH) signaling and body… Show more

“…Pediatric heart disease in general induces GDF15 synthesis and secretion by cardiomyocytes. 33 We detected high GDF-15 and normal FGF-21 levels in nine patients with Gaucher disease (seven children, two adults). The elevation did not correlate with the type of Gaucher disease (three patients had type 2, others had type 1 of Gaucher disease).…”

“…Adenoviruses were generated as previously described (39,40). Transmission electron microscopy, gene expression, Western blot, cellular oxygen consumption measurement, mitochondrial DNA, and enzyme activity analysis were performed as previously described (18,41). Please see SI Appendix, SI Materials and Methods for detailed description of mouse studies, cell culture, glucose uptake, histological analysis, RNA-Seq, ChIP-Seq, and ChIP-reChIP-Seq experiments.…”

“…In physiological states GDF-15 is secreted by cardiac myocytes and it regulated growth and proliferation of tissues located in heart and vessels [10]. Therefore, GDF-15 is realized by cardiac cells due to fluid retention and pressure overload, as well as it produced in resulting in inflammation and cell-to-cell cooperation [11].…”
