Saussurea lappa extract suppresses TPA-induced cell invasion via inhibition of NF-κB-dependent MMP-9 expression in MCF-7 breast cancer cells

Abstract: BackgroundSaussurea lappa (SL) has been used as a traditional herbal medicine to treat abdominal pain and tenesmus, and has been suggested to possess various biological activities, including anti-tumor, anti-ulcer, anti-inflammatory, anti-viral, and cardiotonic activities. The effect of SL on breast cancer metastasis, however, is unknown. Cell migration and invasion are crucial in neoplastic metastasis. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix, is a major component in cancer … Show more

“…MMPs are a major group of enzymes, zinc- and calcium-dependent endopeptidases, that regulate the ECM composition ( Kim et al ., 2014 ). MMP-9 in particular, considered to be one of the critical MMPs involved in cancer invasion, has been found to be directly associated with breast cancer invasion, metastasis, and poor prognosis ( Brinckerhoff and Matrisian, 2002 ).…”

“…Results demonstrated that the functional vincristine plus dasatinib liposomes exhibited the most significant inhibitory effect to cancer cell invasion as compared to the control formulations. The mechanism could be related to the regulation of MMPs in the extracellular matrix [13]. It is known that the secreted MMP-2 and MMP-9 in cancer cells cause hydrolysis of the extracellular matrix, enabling cancer cells to break through the original site into circulation system and finally form the secondary metastatic sites [38].…”

“…Matrix metalloproteinases (MMPs), a family of zinc-dependent endopeptidases, play a major role in remodeling of extracellular matrix structures [12]. Among the identified MMPs, the activity of MMP-2 and MMP-9 dominated the degradation of basement membranes [13,14]. …”

“…Saussurea lappa has traditionally been used in Asian countries as an ingredient in the treatment of abdominal pain and tenesmus, and an analgesic, digestive aid, aphrodisiac, and diuretic. Many authors have reported that S. lappa exhibits biological effects such as antiulcer, anticancer, and hepatoprotective effects ( Byambaragchaa et al, 2014 ; Chen et al, 1995 ; Kim et al, 2014 ; Yoshikawa et al, 1993 ). We have recently reported anti-AD effects of S. lappa as shown in both in vitro and in vivo assays ( Lim et al, 2014 ).…”
