Roles of Podoplanin in Malignant Progression of Tumor

Abstract: Podoplanin (PDPN) is a cell-surface mucin-like glycoprotein that plays a critical role in tumor development and normal development of the lung, kidney, and lymphatic vascular systems. PDPN is overexpressed in several tumors and is involved in their malignancy. PDPN induces platelet aggregation through binding to platelet receptor C-type lectin-like receptor 2. Furthermore, PDPN modulates signal transductions that regulate cell proliferation, differentiation, migration, invasion, epithelial-to-mesenchymal trans… Show more

“…We previously established cancer−specific mAbs (CasMabs) targeting to PDPN [ 34 , 35 , 36 , 37 ] and podocalyxin [ 38 ], which are expressed on many cancers, including ESCC [ 39 , 40 , 41 , 42 ]. These CasMabs recognize cancer−specific aberrant glycosylation of the target proteins [ 43 ]. It is worthwhile to develop a cancer−specific anti−CD44 antibody using the CasMab method.…”

“…Moreover, it has been speculated that CLEC-2 depletion did not show severe bleeding tendency, so that to some extent targeting platelet CLEC-2 is a safe treatment strategy (43,46,48). In addition, PDPN is also expressed in tumor stroma, including cancer-associated fibroblasts (CAFs) (49). PDPN-positive CAFs exhibit poor clinical outcomes in cancers of the lung (50), breast (51, 52), pancreas (53), and esophagus (54).…”

“…Of the top ten genes by correlation in HNSCC [ 47 ], many were found to have functions that support the known role that CSPG4 plays in cancer pathogenesis, such as cell migration, tissue remodelling and angiogenesis. PDPN encodes a mucin-type TM glycoprotein that has functions in development, immunology, migration, invasion, and metastasis [ 160 , 168 ]. Similarly, PIK3CD, which encodes the delta subunit of PI3K, is a key intracellular mediator of cell growth, proliferation, survival and the EMT [ 169 ].…”
