Role of Y Box Protein-1 in cancer: As potential biomarker and novel therapeutic target

Abstract: The Y-box binding protein (YB-1) is known to be a multifunctional transcription and translation factor during expression of several proteins. It is a vital oncoprotein that regulates cancer cell progression and proliferation. YB-1 is over-expressed in various human cancers such as breast cancer, colon cancer, lung cancer, gastric cancer, oesophageal cancer and glioblastoma. Nuclear expression of YB-1 is found to be associated with multidrug resistance and cancer cell progression. YB-1 is reported to regulate m… Show more

“…YBX1 in the nucleus plays a major role as a transcription factor for various drug resistance‐related genes, including ABCB1 , and in the repair of damaged DNA. However, in the cytoplasm, YBX1 is mainly involved in post‐transcriptional control of several mRNA splices, including epithelial‐to‐mesenchymal transition‐related genes . In the present review, we focus on the oncogenic role of YBX1, its localization in the nucleus, and its association with drug resistance.…”

“…Similarly, carcinogenesis and further malignant progression of cancer are associated with the nine hallmarks of cancer, including cell proliferation, survival, immortalization, inflammation, invasion, metastasis, and others . In addition, most of the hallmarks are closely coupled with enhanced expression of YBX1 …”

“…YB1 promotes SG assembly by controlling translation of an essential SG protein, G3BP1 . In various tumor types including metastatic tumors, YB1 is frequently overexpressed, which correlates with drug resistance and poor prognosis . Blocking SG formation in a mouse tumor implantation model by knocking down YB1 or G3BP1 reduces invasive and metastatic tumors, suggesting that the presence of SGs promotes metastasis .…”
