While carboplatin lacks neurotoxicity, it has considerable myelotoxicity which may lead to ineffective treatment due to lower dose-intensity.…”

“…Toxicity was not similar, however, as CHAP-5 induced neurotoxicity, while CHAC-1 patients had significantly less lasting side effects. When this neurotoxicity is taken into account in calculating TWIST, the median TWIST amounted 22 months in patients receiving CC vs. 10 months, when calculated the same way as published in a previously treated group receiving CAP-5 (8). Thus, while overall survival is similar for both groups, the quality of life remaining is not.…”

“…In order to get an objective measure of the impact of treatment on the patient's quality of life, we calculated TWIST, according to Gelber & Goldhirsch (9). This allowed us to compare the subjective toxicity of this combination with that of the formerly used cisplatinbased CAP-5 schedule as we published earlier (8). Just as in our earlier paper, TWIST was calculated in the following way: all admission days (for laparotomy, fever, blood-or platelet transfusions), every outpatient visit (for cytostatic treatment or blood counts), and all days the patient was bedridden, were added.…”

Of the 45 patients who completed six cycles, 10 suffered from severe neurotoxic side-effects and only 8 were free from neurological symptoms.…”
