Abstract: Transcription of the ~200 mouse and human ribosomal RNA genes (rDNA) by RNA Polymerase I (RPI/PolR1) accounts for 80% of total cellular RNA, around 35% of all nuclear RNA synthesis, and determines the cytoplasmic ribosome complement. It is therefore a major factor controlling cell growth and its misfunction has been implicated in hypertrophic and developmental disorders. Activation of each rDNA repeat requires nucleosome replacement by the architectural multi-HMGbox factor UBTF to create a 15.7 kbp nucleosome … Show more

“…Another form of RNAP I, the RNAP Ia, also possesses catalytic activity but cannot bind to a promoter. SL1 is a protein complex built by TBP and the TAFs TAFIA, TAFIB, TAFIC, and TAFID (Drygin et al, 2010; Tremblay et al, 2022). Then, RRN3 (TIF‐IA) binds to the complex, and RNAP I (RNAP Ib) is recruited to the promoter through interactions of UBTF and SL1 with the DNA‐directed RNA polymerase I subunit RPA49 (POLR1E/PAF53).…”

“…SL1 comprises the subunits TAF1A, TAF1B, TAF1C (homologues of yeast Core Factor subunits), and the two additional factors TAF1D ( 37 ) and TAF12 ( 38 ), and includes the TATA-binding protein (TBP). UBF consists of six consecutive HMG boxes, is a part of initiation complexes ( 39 ) in one splice variant ( 40 ), and binds to the body of actively transcribed rDNA genes ( 41 ), apparently preventing re-association of nucleosomes.…”
