Abstract: BrTet showed significant MDR reversal activity in vitro and in vivo. Its activity may be related to the inhibition of P-gp overexpression and the increase in intracellular accumulation of anticancer drugs. BrTet may be a promising MDR modulator for eventual assessment in the clinic.

“…Importantly, clinical studies indicate that ATP7B expression levels might predict the sensitivity of ovarian and endometrial cancers to cisplatin chemotherapy (Nakayama et al, 2002(Nakayama et al, , 2004Aida et al, 2005). Still, in line with the multifactorial nature of cisplatin efflux (and resistance; see below), small molecules that inhibit specific ABC transporters (for example, the P-glycoprotein-specific inhibitor 5-bromotetrandrine) appear to be unable per se to restore cisplatin accumulation and sensitivity (Jin et al, 2005).…”

“…All measurements were performed in a coded, blinded fashion. The mice were sacrificed at day 33 after drug administration, tumor growth and body weight were determined as previously described by Jin et al (8), and the tumors were resected to measure the intracellular accumulation of DOX by fluorospectrophotometry, to assess the P-gp level by immunohistochemistry, and to detect MDR1 mRNA expression by semi-quantitative RT-PCR analysis.…”

“…16,17 Some reports displayed that one of the important mechanisms of tumorigenesis and resistance to anticancer drugs was blockade of the apoptosis-inducing pathway. 18,19 To reverse MDR and minimize serious undesired side effects of systemic chemotherapy, we have developed a promising DNR/Br Tet-MNPs formulation, and in this study, we focus on its effects on the induction of apoptosis in drug resistant human leukemia K562/A02 cells.…”
