Relative expression of TAp73 and ΔNp73 isoforms

Abstract: The transcription factor p73 belongs to the p53 family of tumour suppressors and similar to other family members, transcribed as different isoforms with opposing pro- and anti-apoptotic functions. Unlike p53, p73 mutations are extremely rare in cancers. Instead, the pro-apoptotic activities of transcriptionally active p73 isoforms are commonly inhibited by over-expression of the dominant negative p73 isoforms. Therefore the relative ratio of different p73 isoforms is critical for the cellular response to a che… Show more

“…Thus, several studies have used the ∆N/TA isoform ratio as a biomarker. The ∆Np73/TAp73 ratio has been associated with clinical response to chemotherapy in hepatocellular carcinoma and various cancer cell lines (13,21).…”

“…3D). By virtue of two transcriptional promoters and complex alternative splicing patterns, p73 occurs in multiple isoforms, at least 24 of which have been described to have pro-apoptotic, anti-apoptotic, or not yet described functions [34,35]. Analysis of p73 expression in RS4;11 cells demonstrated multiple forms of the protein, most of which cannot be identified with certainty in the absence of variantspecific antibodies.…”

“…Although p53 is firmly established to be a tumor suppressor, the role of p73 in human tumorigenesis is not well understood (15,16). The p73 gene contains two promoters and thus encodes the transcriptional domain-containing (TAp73) and the amino deleted (ΔNp73) isoforms (17). TAp73 isoforms contain an amino-terminal transactivation domain and thus can activate the promoters of p53-target genes and induce apoptosis (18,19).…”
