Relationship between the Expression of the Extracellular Matrix Genes SPARC, SPP1, FN1, ITGA5 and ITGAV and Clinicopathological Parameters of Tumor Progression and Colorectal Cancer Dissemination

Abstract: Objective: To evaluate the relationship between the expression of the extracellular matrix (ECM) genes SPARC, SPP1, FN1, ITGA5 and ITGAV and the histopathologic parameters of neoplastic progression and colorectal carcinoma (CRC) dissemination. Methods: A retrospective study was conducted in 114 patients with stage I–IV CRC who underwent primary tumor resection. Quantitative real-time PCR and immunohistochemistry (IHC) assays were performed in samples obtained from the primary tumors. The correlation between th… Show more

“…At +14 d even fewer predicted up-stream regulators (and down-stream genes) were uncovered by IPA but there was an apparent prominent role and activation in OF vs. RE of osteopontin (SPP1) and metallopeptidase inhibitor 1 (TIMP1) both of which are related to induction of ECM formation [63], [64].…”

“…Among them, we identified matrix metalloproteinase 7, adhesion molecule CD44, vitronectin receptor (ITGAV), fractalkine receptor (CX3CR1), and transcription factor FoxC1, which are all known to promote the invasion and metastasis of solid tumors. 25,27,[35][36][37][38][39][40] Intriguingly, the increased expression of ITGAV, FoxC1, and CX3CR1 not only correlated with the elevated c-Jun level but also were enriched in secondary extranodal lymphoma. For personal use only.…”

“…To confirm some of these expression changes we selected several proteins for validation by immunoblotting. ITGAV has been found significantly up-regulated in tumors with positive perineural invasion (p ϭ 0.028) (47) and in breast cancer (48). Viana et al observed a direct relationship between ITGAV and EGFR expression (r ϭ 0.774; p Ͻ 0.001) by IHC, suggesting the possibility of crosstalk in this signaling pathway.…”
