Regulation of CEACAM Family Members by IBD-Associated Triggers in Intestinal Epithelial Cells, Their Correlation to Inflammation and Relevance to IBD Pathogenesis

Abstract: Carcinoembryogenic antigen cellular adhesion molecules (CEACAMs) are intercellular adhesion molecules highly expressed in intestinal epithelial cells. CEACAM1, -3, -5, -6, -7 are altered in patients suffering from colon cancer and inflammatory bowel diseases (IBD), but their role in the onset and pathogenesis of IBD is not well known. Herein, we aim to correlate CEACAM1, -3, -5, -6, -7 expression to the degree of inflammation in pediatric and adult IBD colon biopsies and to examine the regulation of CEACAMs on… Show more

“…Both molecules were recognized to be highly present in human colon epithelia and related to inflammation and tumorigenesis [41]. Defects in CEACAM signalling in intestinal epithelial cells are associated with Inflammatory Bowel Disease (IBD), and even Colitis-Associated Cancer (CAC) [41, 42]. As we revealed the interplay of various cell types including colonocytes and cycling cells in CEACAM1 CEACAM5 interaction in IBD or colorectal cancer patients, it might highlight targeting these cells to reverse the adverse conditions.…”

“…Dead cells were excluded using LIVE/DEAD™ Fixable Dead Cell Stain Kits. Anti–CEACAM1–specific mouse monoclonal antibody (clone number 283340) was shown to be non-cross-reactive with other CEACAM families as described previously ( 15 ). Flow cytometry was performed on a FACS–Canto II (BD Biosciences, Franklin Lakes, NJ, USA) and data were analyzed using FlowJo software (BD Biosciences).…”

“…Western blot was performed as described with some modifications (Saiz-Gonzalo et al, 2021). Briefly, colonic tissue samples were homogenised in tissue lysis buffer (50 mM NaCl, 50 mM NaF, 50 mM Na 4 P 2 O 7 , 5 mM EGTA, 5 mM EDTA, 2 mM Na3VO4, 10 mM HEPES, 1% Triton X-100, pH 7.4) supplemented with 1 × Halt Protease and Phosphatase Inhibitor Cocktail (cat no.…”
