Regulating Acidosis and Relieving Hypoxia by Platelet Membrane-Coated Nanoparticle for Enhancing Tumor Chemotherapy

Abstract: Acidosis and hypoxia of tumor remain a great challenge for cancer therapy. Herein, we developed Hb-LOX-DOX-ZIF8@platelet membrane nanoparticles (H-L-D-Z@PM NPs) to address this problem. Lactate oxidase (LOX) could deplete intratumoral lactate adequately and amplify oxidative stress efficiently. In the meantime, hemoglobin (Hb) was intended to deliver oxygen, relieve hypoxia, and boost the catalytic activity of LOX. The coated PM bestowed active tumor-targeting ability and good biocompatibility to these nanopar… Show more

“…On the other hand, due to the severe hypoxia in TME and the fact that the LOX-related reaction is an oxygen-demanding process, researchers loaded haemoglobin (Hb) into ZIF-8 to deliver oxygen to alleviate the hypoxia in tumour tissues and to enhance the catalytic activity of LOX. 75 ZIF-8, a typical MOF, has been widely used in drug delivery for tumour therapy due to its excellent drug-carrying capacity and acid-sensitive properties. [85][86][87][88] However, the low stability of ZIF-8 in water and potential cytotoxicity limit its application.…”

“…For tumor acidosis and hypoxia localization, Luo et al utilized ZIF‐8 to develop Hb‐LOx‐DOX‐ZIF8@platelet membrane nanoparticles. [ 49 ] LOx sufficiently depleted intratumoral lactate and effectively amplified oxidative stress to make tumors more sensitive to doxorubicin (DOX)‐induced chemotherapy. Meanwhile, hemoglobin (Hb) transported O 2 , relieving hypoxia and enhancing the catalytic activity of LOx.…”
