Opioidrezeptoren (ORs) sind im Gehirn, im Rückenmark und im Verdauungstrakt weit verbreitet und spielen eine wichtige Rolle in der Nozizeption. Alle bekannten ORs sind G-Protein-gekoppelte Rezeptoren (GPCRs) der Familie A. Ein weiteres wichtiges Mitglied dieser großen Proteinfamilie, Rhodopsin, wird durch Licht über eine cis/trans-Isomerisierung eines kovalent gebundenen Chromophors, nämlich Retinal, aktiviert. Wir zeigen nun, wie ein OR mit einem synthetischen Azobenzol-Photoschalter kombiniert werden kann und somit lichtempfindlich wird. Unsere Arbeit erweitert den Anwendungsbereich der Photopharmakologie und umreißt eine generelle Strategie, mit der GPCRs der Familie A, die einen Großteil wichtiger Wirkstofftargets ausmachen, in Photorezeptoren umgewandelt werden kçnnen.
