Ratio Estimates, the Delta Method, and Quantal Response Tests for Increased Carcinogenicity

Abstract: This paper demonstrates the use of the delta method for estimating the variance of ratio statistics derived from animal carcinogenicity experiments. The Cochran-Armitage test (Cochran, 1954, Biometrika 10, 417-451; and Armitage, 1955, Biometrics 11, 375-386) is routinely applied to carcinogenicity data as a test for linear trend in lifetime tumor incidence rates. The computing formula for this test derives from the assumption that the denominators of the quantal response rates are fixed. However, when time-at-… Show more

“…Consequently, the combined incidence of adenomas and carcinomas in females was further analyzed using the continuity-corrected Poly-3 trend test with the Bieler-Williams modification that takes survival into consideration (Bieler and Williams, 1993). The Poly-3 trend test is the default trend test of the National Toxicology Program (NTP), even when survival is not affected as in the case of α-methylstyrene (NTP, 2007).…”

“…The resulting survival-adjusted estimate of π ci is . Bieler and Williams (1993) noted that is a random variable and proposed a corrected variance estimator, which under the assumption that π c1 = π c2 = ⋯ = π ck is given by where , and the plus sign indicates summation over the missing index.…”

“…All reported p values for the survival analyses are two-sided. For neoplasm and nonneoplastic lesion incidences, continuity-corrected Poly-3 tests were used to test for exposure-related trends and pairwise comparisons of each exposure group to the control group, and reported p values are one-sided (Bieler and Williams 1993;Portier and Bailer 1989;Bailer and Portier 1988). 500 ppm 1-BP was significantly less than that of the controls.…”
