Rapid stimulation of a soybean protein-serine kinase that phosphorylates a novel bZIP DNA-binding protein, G/HBF-1, during the induction of early transcription-dependent defenses

Abstract: The G‐box (CACGTG) and H‐box (CCTACC) cis elements function in the activation of phenylpropanoid biosynthetic genes involved in the elaboration of lignin precursors, phytoalexins and the secondary signal salicylic acid as early responses to pathogen attack. We have isolated a soybean cDNA encoding a novel bZIP protein, G/HBF‐1, which binds to both the G‐box and adjacent H‐box in the proximal region of the chalcone synthase chs15 promoter. While G/HBF‐1 transcript and protein levels do not increase during the i… Show more

“…In addition, adjacent G-and H-boxes in the promoter of the common bean chs15 gene are bound by transcription factor G/HBF-1. This protein is phosphorylated upon elicitor treatment (Drö ge-Laser et al, 1997). The presence of these motifs in the PvACCase promoter region suggests coordinated regulation with other genes for phenylpropanoid synthesis.…”

“…gma-MIR-Cand016 and gma-MIR-Cand017 also appear to regulate target genes involved in gene expression. The bZIP transcription factor (TF) seems to be the target of gma-MIR-Cand017; this TF negatively regulates nodulation (Nishimura et al, 2002) and is present in plant-defense responses (Dröge-Laser et al, 1997). Other transcription factors have been shown to be regulated by miRNAs during nodulation, such as MtHAP2, which is controlled by miR169; HD-ZIPIII, regulated by miR166; and NAC1, regulated by miR164; all were identified during nodulation in M. truncatula (Combier et al, 2006;Boualem et al, 2008;D'haeseleer et al, 2011).…”

“…For example, phosphorylation of Gbox binding factor 1 from Arabidopsis or of H/G-box binding factor 1 in elicited soybean cells increases the DNA binding activities of these factors (16,17). On the other hand, phosphorylation of opaque 2 from maize causes the opposite effect (18).…”
