Radiolabeled cetuximab: Dose optimization for epidermal growth factor receptor imaging in a head‐and‐neck squamous cell carcinoma model

Abstract: Noninvasive imaging of the epidermal growth factor receptor (EGFR) in head‐and‐neck squamous cell carcinoma could be of value to select patients for EGFR‐targeted therapy. We assessed dose optimization of 111Indium‐DTPA‐cetuximab (111In‐cetuximab) for EGFR imaging in a head‐and‐neck squamous cell carcinoma xenograft model. 111In‐cetuximab slowly internalized into FaDu cells in vitro, amounting to 1.0 × 104 molecules cetuximab per cell after 24 hr (15.8% of added activity). In nude mice with subcutaneous FaDu x… Show more

“…Cetuximab and cetuximab-F(ab9) 2 were conjugated to isothiocyanatobenzyl-diethylenetriaminepentaacetic acid (ITC-DTPA; Macrocyclics) in 0.1 M NaHCO 3 , pH 9.5, for 1 h at room temperature using a 50-fold molar excess of ITC-DTPA (14,15). Unbound ITC-DTPA was removed by dialysis against NH 4 OAc (0.25 M, pH 5.5).…”

“…Previously, we have labeled cetuximab with 111 In and imaged the antibody uptake with SPECT in nude mice bearing FaDu squamous cell carcinoma xenografts (14). The FaDu tumors were clearly visualized.…”

“…25,26 In brief, antibodies were conjugated with isothiocyanatobenzyl-diethylenetriaminepentaacetic acid (ITC-DTPA, Macrocyclis, Dallas, TX) in 0.1 M NaHCO 3 , pH 9.5, using a 50-fold (cetuximab) or 15-fold (R1507 and hLL2) molar excess of ITC-DTPA for 1 hr at room temperature. The unbound ITC-DTPA was removed from the reaction mixture by dialysis against 0.25 M ammonium acetate buffer, pH 5.4.…”

“…While whole antibodies accumulate efficiently in the tumor, it requires 4-7 days before non-target radioactivity has reached sufficiently low levels to enable acquisition of high contrast SPECT images (10). Conversely, F(ab′) 2 fragments demonstrate high tumor-to-background signal ratios at earlier time points, while retaining tumor-binding characteristics similar to those of whole antibodies (11,12).…”
