Quantification of phenylalanine hydroxylase activity by isotope-dilution liquid chromatography–electrospray ionization tandem mass spectrometry

Abstract: BACKGROUND: Residual phenylalanine hydroxylase (PAH) activity is the key determinant for the phenotype severity in phenylketonuria (PKU) patients and correlates with the patient's genotype. Activity of in vitro expressed mutant PAH may predict the patient's phenotype and response to tetrahydrobiopterin (BH(4)), the cofactor of PAH. METHODS: A robust LC-ESI-MSMS PAH assay for the quantification of phenylalanine and tyrosine was developed. We measured PAH activity a) of the PAH mutations p.Y417C, p.I65T, p.R261Q… Show more

“…7,9 A number of mutations are more frequently found in BH4 responders, whereas BH4 nonresponders harbor more severe mutations, with less or little residual PAH activity. 38 All of the 85 genotypes identified within our survey and the 8 mutations detected on only 1 allele were reported to be associated with substantial (.30%) residual PAH activity (www.biopku.org).…”

“…In mice, Pah is mainly expressed in liver but there is also considerable expression in kidney 29 ( Supplementary Figure S3 ). We analyzed Pah -mRNA, as well as PAH protein and enzyme activity in Pah enu2/+ mice subjected to different treatment regimes.…”

“…PAH enzyme activity in liver extracts was measured according to a highly sensitive and quantitative assay using isotope-dilution liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MSMS) as described. 29 Briefly, assay conditions included preincubation at 25 °C with L-Phe (1 mmol/l) for 4 minutes, then Fe(NH 4 ) 2 (SO 4 ) 2 (100 ìmol/l) was added, and incubation was continued for one more min. After 5 minutes total preincubation time, BH 4 (75 ìmol/l) was added to start the reaction.…”

“…PAH activities were determined using a novel mass spectrometry method for quantification of Phe and Tyr in cell lysates [19]. Mutations in the human PAH cDNA sequence in pCMV-FLAG-PAH were introduced by site-directed mutagenesis using QuikChange XL II kit from Agilent Technologies (Santa Clara, CA, USA) and confirmed by DNA sequence analysis.…”
