Abstract: Intermittent fasting (IF) increases lifespan and decreases metabolic disease phenotypes and cancer risk in model organisms, but the health benefits of IF in humans are less clear. Human plasma derived from clinical trials is one of the most difficult sample sets to analyze using mass spectrometry-based proteomics due to the extensive sample preparation required and the need to process many samples to achieve statistical significance. Here, we describe an optimized and accessible device (Spin96) to accommodate … Show more

“…Samples were then processed for trypsin digestion and cleanup using the SP3 method (Hughes et al, 2014). After SP3 peptide eluates were additionally cleaned using SDB-RPS StageTips as described previously (Harney et al, 2019b). Peptides were resuspended in 5% formic acid and stored at 4 C until acquired by LC-MS.…”

“…Mouse Plasma Proteomics Confirms a1-Antitrypsin Downregulation during EODF Given our surprising finding that the abundant blood plasma protein a1-antitrypsin (SERPINA1E) was significantly decreased in the liver of animals on the EODF diet regime, we wanted to determine if the regime also induced a similar change in the blood plasma proteome. To achieve this, we used undepleted plasma proteomics (Harney et al, 2019b), to process all plasma samples for trypsin digestion prior to LC-MS/MS analysis. This yielded the identification of 170 protein groups (Table S5) from a total of >7,500 peptides (Table S6).…”

“…The textured surface of simple 3‐D‐printed devices may be undesired in a working prototype, or the material choice may be suboptimal to address the functional requirements of a device. Examples of 3‐D‐printed devices have been presented for use in the proteomics lab, although are often used to generate components of more complex devices (such as a holder). Realize that 3‐D printing is simply a means to manufacturing—printed devices are not necessarily better than those built by other means and may not even be cheaper or simpler to produce.…”

“…It is defined as abstinence or reduction of food, drink, or both for a period lasting typically between 12hours (hrs) and 3 weeks, in short-term, long-term or an intermittent pattern [4,5]. Intermittent fasting is an umbrella term for various diets that cycle between a period of fasting and non-fasting [6]. It is gaining momentum as a strategy to improve human health.…”
