Protective effects ofOriganum vulgareethanol extract against cyclophosphamide-induced liver toxicity in mice

Abstract: Context: Despite its wide clinical use, cyclophosphamide (CP), an alkylating chemotherapeutic agent, possesses many adverse effects, including hepatotoxicity. Because Origanum vulgare L. (Lamiaceae) has antioxidative properties, it might protect against above-mentioned damage. Objective: This study evaluated the protective effects of O. vulgare extract on CP-induced liver toxicity. Materials and methods: Mice were pretreated with aerial parts of O. vulgare ethanolic extract (intraperitoneally) at doses of 50, 1… Show more

“…Furthermore, no increase in serum concentration of ALT and AST may provide evidence to protect of liver against hepatocellular degeneration (Al-Jaff, 2011). The presented study confirmed previous results (Habibi, et al, 2014;Saleh, 2014;Zhu, et al, 2014) on the decreasing effect of phytoadditives on the activities of plasma ALT and AST in animals. Meanwhile, Abd ElGhang and Ismail (2013) who used oregano essential oil in broiler feed, observed increase in the activity of ALT and AST.…”

“…Cyclophosphamide also undergoes an inactivation pathway (to form the metabolite DECP) mediated by CYP3A13 in mice (Figure a; Pass et al, ). Treatment with cyclophosphamide (i.p., 300 mg·kg −1 , n = 5) induced hepatotoxicity in wild‐type mice (Figure b) consistent with previous reports (Habibi et al, ; Sheweita, El‐Hosseiny, & Nashashibi, ). Of note, the severity of hepatotoxicity was dosing time dependent with high levels at ZT2/22 and low levels at ZT10/14 (Figure b).…”

“…Furthermore, no increase in serum concentration of ALT and AST may provide evidence to protect of liver against hepatocellular degeneration (Al-Jaff, 2011). Presented experiment did not confirm results of other authors (Habibi et al, 2014;Saleh, 2014;Zhu et al, 2014). Shi et al (2014) who added phytosterols into feed mixture of layers reported some minor changes in clinical chemistry parameters, but these changes were small and considered to be of no toxicological significance.…”
