Proneural gene-linked neurogenesis in zebrafish cerebellum

Abstract: In mammals, cerebellar neurons are categorized as glutamatergic or GABAergic, and are derived from progenitors that express the proneural genes atoh1 or ptf1a, respectively. In zebrafish, three atoh1 genes, atoh1a, atoh1b, and atoh1c, are expressed in overlapping but distinct expression domains in the upper rhombic lip (URL): ptf1a is expressed exclusively in the ventricular zone (VZ). Using transgenic lines expressing fluorescent proteins under the control of the regulatory elements of atoh1a and ptf1a, we tr… Show more

“…We sought to determine which cell types respond to injury and contribute to cellular regeneration. The transgenes ptf1a:DsRed and nestin:gfp distinctly label VZ progenitor cells and neuroepithelial-like progenitor cells, respectively, in the zebrafish cerebellum, and they can be used to monitor stem and progenitor cell activity (Kani et al, 2010;Kaslin et al, 2013). Furthermore, Ptf1a is required for the generation of all inhibitory neuronal lineages in the cerebellum (Hoshino et al, 2005).…”

“…Although the presence of an EGL in anamniotes has been debated in recent years (Wullimann et al, 2011), there is little evidence for its manifestation in the cerebellum of sharks (Rodriguez-Moldes et al, 2008;Chaplin et al, 2010), basal ray-finned fish (Butts et al, 2014) and early or adult zebrafish (Kaslin et al, 2009(Kaslin et al, , 2013Chaplin et al, 2010;Kani et al, 2010). We conclude that a transit amplifying precursor layer is also absent in the frog.…”

“…Microinjection of Tol2-based plasmid DNA was performed as described previously (Kawakami, 2004;Urasaki et al, 2006 (Miyasaka et al, 2009;Kani et al, 2010;Kinkhabwala et al, 2011;Koyama et al, 2011) Stochastic labeling of V0 neurons with GFP. For stochastic expression of GFP in V0 neurons, gad1b:lDl-GFP, or huC:lDl-GFP was injected into one to four cell stage embryos of Tg[dbx1b:Cre] stable transgenic fish.…”
