Since its identification, 75 years ago, the monoamine serotonin (5-HT) has attracted considerable attention toward its role as a neurotransmitter in the central nervous system. Yet, increasing evidence, from a growing number of research groups, substantiates the fact that 5-HT regulates important nonneuronal functions. Peripheral 5-HT, synthesized by the enzyme tryptophan hydroxyase (Tph) in intestinal cells, was assumed to be distributed throughout the entire body by blood platelets and to behave as a pleiotropic hormone. A decade ago, generation of a mouse model devoid of peripheral 5-HT lead to the discovery of a second isoform of the enzyme Tph and also suggested that 5-HT might act as a local regulator in various organs. The objective of this review is to highlight the newly discovered functions played by the monoamine using the Tph1 KO murine model and to outline current findings that led to the discovery of complete serotonergic systems in unexpected organs. Within an organ, both the presence of local Tph enzymatic activity and serotonergic components are of particular importance as they support the view that 5-HT meets the criteria to be qualified as a monoamine with a paracrine/autocrine function.

BackgroundPlatelet-derived growth factor is involved in the regulation of hematopoiesis. Imatinib mesylate, a platelet-derived growth factor receptor inhibitor, induces thrombocytopenia in a significant proportion of patients with chronic myeloid leukemia. Although our previous studies showed that platelet-derived growth factor enhances megakaryocytopoiesis in vitro, the in vivo effect of platelet-derived growth factor in a model of radiation-induced thrombocytopenia has not been reported. Design and MethodsIn this study, we investigated the effect of platelet-derived growth factor on hematopoietic stem/progenitor cells and platelet production using an irradiated-mouse model. We also explored the potential molecular mechanisms of platelet-derived growth factor on thrombopoiesis in M-07e cells. ResultsPlatelet-derived growth factor, like thrombopoietin, significantly promoted the recovery of platelets and the formation of bone marrow colony-forming unit-megakaryocyte in irradiated mice. Histology confirmed the protective effect of platelet-derived growth factor, as shown by an increased number of hematopoietic stem/progenitor cells and a reduction of apoptosis. In a megakaryocytic apoptotic model, platelet-derived growth factor had a similar anti-apoptotic effect as thrombopoietin on megakaryocytes. We also demonstrated that platelet-derived growth factor activated the PI3-k/Akt signaling pathway, while addition of imatinib mesylate reduced p-Akt expression. ConclusionsOur findings show that platelet-derived growth factor enhances platelet recovery in mice with radiation-induced thrombocytopenia. This radioprotective effect is likely to be mediated via platelet-derived growth factor receptors with subsequent activation of the PI3-k/Akt pathway. We also provide a possible explanation that blockage of platelet-derived growth factor receptors may reduce thrombopoiesis and play a role in imatinib mesylate-induced thrombocytopenia.Key words: platelet-derived growth factor, thrombopoiesis, imatinib mesylate, thrombocytopenia, apoptosis. Mögliche Anwendungen von Stammzellen in der Medizin reichen von der Modellierung von Krankheiten und der Wirkstoffsuche bis hin zu Zelltransplantation und regenerativen Therapien. Bevor diese Versprechen jedoch eingelöst werden können, müssen noch einige Hindernisse überwunden werden, unter anderem die Kontrolle der Stammzelldifferenzierung, die allogene Abstoßung und die eingeschränkte Zellverfügbarkeit. Dies erfordert ein vertieftes Verständnis der Mechanismen, die das Stammzellpotenzial kontrollieren, und die Entwicklung robuster Methoden, um das Schicksal von Stammzellen effizient zu steuern. In der letzten Zeit wurden eine Reihe niedermolekularer Verbindungen entdeckt, die in vitro und in vivo verwendet werden können, um Stammzellen zu expandieren, ihre Differenzierung zu dirigieren oder somatische Zellen in ein naiveres Stadium zu reprogrammieren. Diese Moleküle haben tiefe Einblicke in Signalwege und epigenetische Mechanismen ermöglicht, die die Stammzellbiologie regulieren, und sie beginnen bereits, zur Entwicklung effizienter Behandlungen für Gewebereparatur und –regeneration beizutragen.
