Promoting effect of 1,25(OH) 2 vitamin D 3 in osteogenic differentiation from induced pluripotent stem cells to osteocyte-like cells

Abstract: We recently reported a new method to purify the induced pluripotent stem (iPS)-derived osteoprogenitors (iPSop). In this paper, we optimized the procedure and characterized cells at each process step. We observed that 10 days of treatment with FGF-2, IGF-1 and TGF-β (FIT) resulted in early-phase osteoblasts and 14 days of treatment resulted in late-phase osteoblasts. We found that treatment with 1,25(OH)2 vitamin D3 increased expression of osteocalcin and decreased expression of tissue-non-specific alkaline ph… Show more

“…Vitamin D3 can be alternately obtained from the diet or in the form of Vit D3 (derived from animals) or vitamin D2 (derived from plants). From the biological aspects, Vit D3 greatly influences the bone cell and participates in the regulation of osteoblasts and osteoclasts activities which influence the processes of bone synthesis, resorption, and regeneration . Bone regeneration is a dynamic process which is orchestrated by osteoblast and osteoclast cells.…”

“…Links between vitamin D and bone metabolism have been reported in the literature, and its effect on stimulating osteoblast proliferation and differentiation (Kato et al. ), but the effect of the topical application of osteoinductive elements like vitamin D over immediate implants is still subject of investigation.…”

“…Experiments suggest also application of active form of 1,25(OH)2 D3 vitamin as the induction factor of osteogenesis (Kato et al 2015). It was shown that there is possibility of generation both the osteoblasts and the cells similar to osteocytes from induced pluripotent cells inter alia by the supplementation with 1,25(OH)2 D3 vitamin (Kato et al 2015).…”
