Promoter-Specific Hypomethylation Is Associated with Overexpression of PLS3 , GATA6 , and TWIST1 in the Sezary Syndrome

Abstract: The Sézary Syndrome (SS) is an aggressive CD4+ leukemic variant of cutaneous T-cell lymphoma. Epigenetic modification of cancer cell genome is often linked to the expression of important cancer-related genes. Here we addressed the hypothesis that, in SS, DNA hypomethylation is involved in upregulation of PLS3, GATA6, and TWIST1, genes that are undetected in normal lymphocytes. Pyrosequencing analysis of CpG rich regions, and CpG dinucleotides within the 5' regulatory regions, confirmed hypomethylation of all t… Show more

“…PLS3 is a kind of plastin protein expressed in a variety of organs, which is associated with various diseases (Sugimachi et al., ; Wong et al., ; Yanyan et al., ). The protein mainly functions in the dynamic assembly and disassembly of actin bundles (Tanaka‐Kamioka, Kamioka, Ris, & Lim, ).…”

“…Analyses of the T cells in Sz patients and normal controls revealed particularly high selective expression of EphA4 and TWIST as well as in other types of CTCL (van Doorn et al, 2004). More recently, promoter DNA hypomethylation of TWIST has been associated with protein overexpression (Wong et al, 2015).…”

“…It is widely accepted that promoter hypermethylation causes a downregulation of gene expression, whereas promoter hypomethylation is associated with the upregulation of gene expression1920. To understand if changes in promoter methylation affect gene transcription in PTC, we evaluated groups of genes that showed both differential methylation and expression between tumor and normal tissues.…”
