Background: Histidine-rich glycoprotein (HRG), a multifunctional plasma protein, has a regulatory role in homeostasis, angiogenesis, and immunity; which in turn could greatly affect tumor control and metastasis. Objectives: To assess the possible role of HRG in acute lymphoblastic leukemia (ALL) tumorgenesis and follow-up. Design and Methods: HRG was quantitatively measured in serum by ELISA and its expression was assessed by real-time PCR (qPCR) in 35 patients with ALL and compared to same 25 ALL patients after induction therapy and 30 age and sex matched healthy control subjects. Results: HRG-serum protein (at cutoff value 63.55 pg/ml) and HRG-RNA (at cutoff value 0.955) were positive in all ALL patients before therapy, but in only 76% after therapy for HRG-protein and 60% for HRG-RNA and they could not be detected in the control group; P < 0.001. Additionally, the serum HRG level showed a significant positive correlation with its expression level, bone marrow blast percentage, peripheral blood blast count, P < 0.01. Also its serum and expression levels were positively related to the poor risk Philadelphia chromosome; P < 0.01. Conclusions: HRG (protein and RNA) might be considered as a novel diagnostic and prognostic marker in ALL. HRG-serum protein level, detected by simple methodology of ELISA, has more significant advantages than its expression level, motivating its application in large clinical studies as a potential marker.

Uvod: Survivin (BIRC5) je pripadnik familije proteina inhibitora apoptoze i ima izuzetno važnu ulogu u takozvanoj tački bez povratka ćelijske apoptoze. Već je dokazana uloga survivina u nastanku tumora i autoimunih oboljenja, međutim sve je više dokaza da je takođe senzitivan na postojanje prethodnog ishemijskog oboljenja srca. Metode: U ovu prospektivnu pilot studiju je uključeno 78 pacijenata koji su se pripremali za jednu od opsežnih nekardiohirurških operacija. Krv je uzorkovana unutar 7 preoperativnih dana, izdvojen je serum i zamrznut na-70оC. Merenje količine survivina u serumu je sprovedeno ELISA (Enzyme-linked immunosorbent assay) metodom, a rezultati su očitavani na aparatu DIAREADER Elx800G. Rezultati: Pacijenti su bili prosečne starosti 71,46 ± 6,87 godina, dok je medijana survivina iznosila 4,56 (0,00-76,78) pg/ml. Survivin se pokazao kao odličan prediktor postoperativne smrtnosti (P < 0,05, ROC/AUC = 0,807). ObjectivesBaicalein is a Chinese traditional medicine that inhibits tumor migration and growth. Pancreatic neuroendocrine tumors (pNETs) have a high incidence in China, but there are still no effective treatments. The aim of our study was to investigate whether baicalein could inhibit pNETs.MethodsIn vitro, we used BON1—a cell line of pNETs—to analyze the apoptosis and migration and invasion after baicalein treatment via flow cytometry and Western blot. In vivo, we used a xenograft tumors model to evaluate the size of tumors after baicalein treatment. Western blot was used to analyze the expression of apoptosis and migration-related protein.ResultsIn vitro, the Cell Counting Kit 8 assay showed that baicalein decreased BON1 viability, and flow cytometry demonstrated that baicalein induced BON1 apoptosis and protein changes. In addition, baicalein inhibited BON1 migration and invasion as shown via a Transwell assay. In vivo, baicalein inhibited tumor growth and migration and also increased apoptosis-related protein expression.ConclusionsBaicalein could increase caspase-3 and Bax expression and decrease survivin and Bcl-2 to induce apoptosis. It inhibits migration and invasion by decreasing expression of vascular endothelial growth factor and matrix metalloproteinases 2 and 9.
