Production of serum antibodies that recognise epitopes located on the R3 region of Escherichia coli core lipopolysaccharides by patients infected with strains of enterohaemorrhagic E. coli

Abstract: Antibody-antigen cross-reactions were examined with sera from patients with Escherichia coli O157 infection and lipopolysaccharide (LPS) purified from a range of enterohaemorrhagic E. coli (EHEC) including those belonging to serogroups O26, O103, O111, O145 and O157. Six of 10 patients infected with an O157 EHEC produced serum antibodies that cross-reacted with common LPS-core epitopes, which were expressed by 23 of 33 strains of EHEC examined. These common LPS-core epitopes were also present on strains of E. … Show more

“…5 Some patients infected with EHEC strains may produce antibodies reactive with the R3-core of the LPS molecule that is present in the majority of EHEC and other E. coli strains. 27,28 The cross-reactivity between the various LPS was evidenced by the absorption assays, even in the absorption with LPS O111 and LPS O26 that were selected due to the presence of the R3-core in these LPS. 27 The reductions in anti-O55 and anti-O157 LPS IgM levels may be attributed to the absorption of antibodies reactive to common epitopes of these LPS.…”

“…The J5 LPS has been widely used in immunochemical studies, in analyses of endotoxicity, and in attempts to develop antibodies against core OS-lipid A that would be broadly cross-reactive and protective in applications such as treatment of sepsis (15)(16)(17)(18)(19). Antibodies cross-reacting with R3 core OS epitopes were detected in 6 of 10 patients infected with different EHEC strains (20), and it has been proposed that IgG and IgA responses to the R3 core OS may be protective against E. coli and Shigella spp. (21).…”

“…2001). A study by Chart et al. (2002) used human serum antibodies, from patients infected with strains of EHEC, to demonstrate that routine serodiagnosis of infections with O157 VTEC was not influenced by antibodies to the R3‐core.…”
