Previous studies have demonstrated that cocoa intake decreased Th2 immune-related antibodies in rats. In consequence, we aimed to study in depth this cocoa action, particularly assessing its effect on a rat model of food allergy (FA) and also on an anaphylactic response. The involvement of the intestinal immune system was analyzed to allow the action mechanisms to be investigated. The role of cocoa flavonoids in the antiallergic properties of cocoa was also established. Brown Norway rats were fed either a reference diet or diets containing conventional cocoa (CC) or nonfermented cocoa (NFC). FA to ovalbumin (OVA) was induced and, later, an anaphylactic response was provoked. As expected, the synthesis of anti-OVA IgE and other Th2-related antibodies was inhibited by CC diet. In addition, the release of mast cell protease II after anaphylaxis was partially prevented by CC, although other variables were not modified. The CC diet also attenuated the increase of some Th2-related cytokines released from mesenteric lymph node and spleen cells, and modulated the intestinal gene expression of molecules involved in allergic response. These results demonstrated the local and systemic influence of CC diet. The effects of the NFC diet were weaker than those of CC, suggesting that cocoa components other than flavonoids play a role in cocoa's action. In conclusion, by acting on intestinal and systemic immune functions, a cocoa-enriched diet in rats exhibited a protective effect against FA and partially against anaphylaxis, making this a food of high interest to the fields of health and immunonutrition.

Abstract. Cancer metastasis is one of the most critical events in cancer patients, and the median overall survival of stage IIIb or IV patients with metastatic lung cancer in the TNM classification is only 8 or 5 months, respectively. We previously demonstrated that Juzentaihoto, a Japanese traditional medicine, can inhibit cancer metastasis through the activation of macrophages and T cells in mouse cancer metastatic models; however, the mechanism(s) through which Juzentaihoto directly affects tumor cells during the metastasis process and which herbal components from Juzentaihoto inhibit the metastatic potential have not been elucidated. In this study, we focused on the epithelial-to-mesenchymal transition (EMT), which plays an important role in the formation of cancer metastasis. We newly determined that only the Cinnamomi Cortex (CC) extract, one of 10 herbal components of Juzentaihoto, inhibits TGF-β-induced EMT. Moreover, the contents of catechin trimer in CC extracts were significantly correlated with the efficacy of inhibiting TGF-β-induced EMT. Finally, the structure of the catechin trimer from CC extract was chemically identified as procyanidin C1 and the compound showed inhibitory activity against TGF-β-induced EMT. This illustrates that procyanidin C1 is the main active compound in the CC extract responsible for EMT inhibition and that procyanidin C1 could be useful as a lead compound to develop inhibitors of cancer metastasis and other diseases related to EMT.

Abstract.To evaluate whether an aqueous seed extract of Terminalia chebula Retzius inhibited development of atopy in vivo, we used a 2,4-dinitrofluorobenzene (DNFB)-induced animal model of atopic symptoms to investigate the effects of the extract. We measured CD4 + cell numbers by hematoxylin and eosin (H&E) staining, and determined the expression levels of matrix metalloproteinase (MMP)-9, interleukin (IL)-31, and T-bet genes, in this animal model. The data showed that a Terminalia chebula extract (100 µg/ml) exhibited strong anti-atopic activity, mediating a 52% reduction in the immune response, as measured by thickness of ear swelling, and resulting in decreased eosinophil levels in adjacent skin tissue. Collectively, the results indicate that a Terminalia chebula seed extract has potential for alleviation of atopy-like symptoms induced by DNFB in the mouse. IntroductionIt is generally accepted that the daily consumption of plantderived phytochemicals in vegetables, fruit, tea or herbal extracts may mitigate free-radical attacks (1-3). Terminalia chebula Retzius is an ethnopharmacological plant of India and Southeast Asia, and has traditionally been used as a laxative, a diuretic, and an anti-oxidative material (4-8). In addition, plant extracts have antibacterial (9,10), antiviral (11), antifungal (12) and immune modulatory activities (13,14). Recently, Lee et al (5,15) reported that an aqueous seed extract of Terminalia chebula Retzius (TcRSE) had potent anti-oxidant activity both in vitro and in vivo when tested as a hepatoprotective agent in an animal model. Atopic dermatitis is a major degenerative disease, which presents as a chronic inflammatory skin condition with a deficiency in barrier function (16). It is now well recognized that several proteases, including the matrix metalloproteinases (MMPs), play key roles in the immunopathology of and biological functions during progression of the disease (17). With limited exceptions, normal healthy skin does not contain or exhibit high levels of MMP activity. Thus, evaluation of enzyme activities is important in detection, prior to mitigation, of allergic reactions (18,19). However, little effort has been devoted to treatment of atopy-related disorders with natural extracts, and we thus prioritized the investigation of TcRSE as an inhibitor of anti-atopic dermatitis activity.In the present study, we initially sought immunomodulatory components in natural resources including foods and oriental herbs, and found that TcRSE was active in vitro (data not shown).
