Pressure simulation of orthodontic force in osteoblasts: a pilot study

Abstract: In mature osteoblasts centrifugal pressure force stimulates RUNX2 gene expression within a narrow time frame: loading of mature cells results in a temporary increase of RUNX2 expression and a fast downregulation back to its pre-load expression level. With this pilot study the gene expression behavior after mechanical stimuli could be determined with a simple laboratory setup.

“…Consider F = 1.18 × 10 −5 × R × RS 2 (Figure 1(a)). Then SCAPs were cultured in the plates and centrifuged at 200 g (corresponded to a speed at 1000 rpm) for 30 min [15], which resembles the clinical orthodontic force, and then returned to the incubator. Control cells were cultured in absence of centrifugal loading.…”

“…where P = kg pressure per cm 2 of cells, m = mass of medium (0.002 kg), r = radius (0.09 m), rpm = revolution /min (600 or 4,800) and A = area of contact between medium and cells (9.62 cm 2 ) (20)(21)(22)(23). The cells on the dishes were then cultured continuously up to 7 days.…”

“…In contrast, the former study by Chang et al did not show an increase in proteins that contribute to ECM formation. Similarly, mechanical stimuli have been shown to promote cellular responses in the form of centrifugal force induced RUNX2 expression in osteoblasts [Baumert et al, 2004], mechanical strain of SaOS-2 cells in vitro affects expression of collagen genes types I and III [Liu et al, 2005], and changes in the mechanical environment such as simulated microgravity or substrate deformation have been shown to affect cellular behavior [Granet et al, 2001[Granet et al, , 2002.…”
