Preparation of multiple-unit floating-bioadhesive cooperative minitablets for improving the oral bioavailability of famotidine in rats

Abstract: The aims of this study were to prepare fine famotidine-containing floating-bioadhesive cooperative minitablets and to investigate the possibility of using those minitablets as a delivery system for promoting the oral bioavailability of famotidine. Nine minitablet formulations were designed using hydroxypropylmethylcellulose (HPMC K4M) as releaseretarding polymers, Carbopol 971P as bioadhesive materials and sodium bicarbonate (NaHCO 3 ) as gas formers. The prepared 3 ± 0.02 mm minitablets were evaluated in term… Show more

“…Over the past few decades, various approaches have been proposed to realize gastro retention, such as floating drug delivery systems (FDDSs), mucoadhesive drug delivery systems, swellable drug delivery systems and other delayed gastric emptying devices [7]. FDDSs could basically achieve floating in the gastric fluid because of its lower bulk density compared with that of the aqueous medium.…”

“…[6] Over the past few decades, various approaches have been proposed to realize gastro retention, such as floating drug delivery systems (FDDSs), mucoadhesive drug delivery systems, swellable drug delivery systems and other delayed gastric emptying devices. [7] FDDSs could basically achieve floating in the gastric fluid because of its lower bulk density compared with that of the aqueous medium. Among them, multiple-unit floating pellets are given on more emphasis because of their various advantages over single-unit floating tablets demonstrated as flexibility during formulation development and therapeutic benefits for the patients.…”

“…The most important characteristic of MUDFs in comparison to SUDFs is their lower susceptibility to dose dumping and to a faster gastric emptying. This is because the subunits of a MUDF may be distributed more evenly throughout the gastro intestinal tract [12,13]. Such advantages result in fewer adverse effects, better bioavailability, lower variability in the drug release and, consequently, better compliance by patients [14].…”
