Prediagnostic plasma testosterone, sex hormone‐binding globulin, IGF‐I and hepatocellular carcinoma: Etiological factors or risk markers?

Abstract: Elevated prediagnostic testosterone and insulin‐like growth factor I (IGF‐I) concentrations have been proposed to increase risk of hepatocellular carcinoma (HCC). However, the metabolism of these hormones is altered as a consequence of liver damage and they may have clinical utility as HCC risk markers. A case–control study was nested within the European Prospective Investigation into Cancer and Nutrition cohort and included 125 incident HCC cases and 247 individually matched controls. Testosterone, sex hormon… Show more

“…HBV and HCV seropositivity were detected using the ARCHITECT HbsAg and anti‐HCV chemiluminescent microparticle immunoassays (CMIAs; Abbott Diagnostics, France). Circulating IGF‐I levels (ELISA, DSL‐10–2800, Beckman Coulter, Brea, CA) and biochemical markers of hepatic injury have also already been determined in these subjects . The latter include alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma‐glutamyltransferase (GGT), liver‐specific alkaline phosphatase (AP), albumin, total bilirubin and total protein (measured on ARCHITECT c Systems™; Abbott Diagnostics; standard protocols).…”

“…Higher prediagnostic SHBG levels are associated with higher HCC risk. SHBG adds diagnostic benefit to the consideration of liver damage and epidemiological variables alone [66]. Hence, SHBG might constitute a useful biomarker for early detection of HCC.…”

“…Only one prospective study has examined this hypothesis in a population that included females, concluding that sex hormone binding globulin (SHBG), but not testosterone, was associated with increased HCC risk. ( 16 ) However, the study was not powered to examine the associations separately for females.…”
