Potentially important microRNA cluster on chromosome 17p13.1 in primary peritoneal carcinoma

Abstract: MicroRNAs are a group of small non-coding RNAs approximately 22 nucleotides in length. Recent work has shown differential expression of mature microRNAs in human cancers. We characterized the alteration in expression of a select group of microRNAs in primary peritoneal carcinoma relative to matched cases of ovarian serous carcinoma. MicroRNA expression was analysed using semi-quantitative stem-loop RT-PCR on a set of 34 formalin-fixed paraffin-embedded samples. Protein expression of p53 and bcl-2 was quantifie… Show more

“…In oncology reports, miR-195 has been considered as tumor suppressor and/or inhibitor of cell proliferation. miR-195 is down-regulated in tongue squamous cell carcinoma [38], primary peritoneal carcinoma [39], bladder cancer [40], gastric cancer [41], and hepatocellular carcinoma cells [42]. However, in our present study, inhibition of miR-195 itself significantly decreased the cell number, suggesting that the function of this miRNA might depend on the type of cancer cells.…”

“…Introduction of miR-195 markedly suppressed colony formation in vitro and tumor development in nude mice (24). MiR-497 expression was found to be downregulated in different cancers, such as primary peritoneal carcinoma, gastric cancer and breast cancer (23,27). However, the role of miR-497 in human cancer is not clear yet.…”

“…Although, the down-regulation of miR-497 in a primary peritoneal carcinoma was shown [45], nothing is known about the importance of miR-497 in autoimmune disease or immune cells. Target prediction, based on sequence, suggests cadherin, T-cell activation and Wnt-pathway genes as possible targets, but none of them has been experimentally validated.…”
