POFUT1 as a Promising Novel Biomarker of Colorectal Cancer

Abstract: Background: While protein O-fucosyltransferase 1 (POFUT1) overexpression has been recently proposed as a potential biomarker for different cancer types, no study was carried out on POFUT1 implication in colorectal cancer (CRC). Methods: Data from 626 tumors and 51 non-tumor adjacent tissues available in FireBrowse had been used in this study. Statistical analyses on POFUT1 expression and gene copy number, NOTCH receptors (main targets of POFUT1 enzymatic activity) expression and association of POFUT1 and NOTCH… Show more

“…Serum-based biomarkers are of considerable importance in the early diagnosis of various diseases including cancers (27,28). In recent years, the public availability of cancer gene expression data, such as Oncomine databases and TCGA databases, has provided a unique opportunity to explore initial hypotheses and concepts that can subsequently be validated by additional experiments (29)(30)(31). There was much higher APOC1 expression in serum of GC than that in adjacent tissues of GC and normal control.…”

“…A collection of 306 specimens of OSCC was obtained from TCGA database, the level 3 RNA sequence (RNA-seq) data (.count files) was downloaded from TCGA website (https://www.cancer. gov/about-nci/organization/ccg/research/structural-genomics/tcga), and their correlated clinical information from FireBrowse database (http://firebrowse.org/) (Chabanais et al, 2018). The RNA-seq data in the TCGA database was normalized and processed using the "edgR" package (Zhang et al, 2019) prior to log 2 conversion (Yang et al, 2019).…”

“…Gene expression differences included the downregulation of protein O -fucosyltransferase 1 ( POFUT1 ), which is a key factor in the Notch1 ( NOTCH1 ) signaling pathway. This pathway is an important regulator for cell death and has been associated with poorer prognosis (Chabanais et al, 2018). Notch 1 known to be essential for maintenance of normal intestinal epithelium and is activated in primary colorectal cancer (CRC) rather than metastatic colon cancer, it may therefore may be more important for early CRC development (Suman et al, 2014).…”
