Pleiotrophin and metabolic disorders: insights into its role in metabolism

Cristina Ballesteros-Pla,
María Gracia Sánchez-Alonso,
Javier Pizarro-Delgado
et al.

Abstract: Pleiotrophin (PTN) is a cytokine which has been for long studied at the level of the central nervous system, however few studies focus on its role in the peripheral organs. The main aim of this review is to summarize the state of the art of what is known up to date about pleiotrophin and its implications in the main metabolic organs. In summary, pleiotrophin promotes the proliferation of preadipocytes, pancreatic β cells, as well as cells during the mammary gland development. Moreover, this cytokine is importa… Show more

“…Of the genes identified in our internal bulk and single RNA-seq data only PTN, SLC7A8 and CDKN3 were also identified in mice datasets to be similarly expressed. PTN is a cytokine which when knocked out in high-fat diet fed mice, protects against insulin resistance, obesity and neuroinflammation Acharjee et al Journal of Translational Medicine (2024) 22:592 [52]. Similarly, deletion of SLC7A8 in mice offers significant protection against diet-induced obesity and enhances glucose metabolism.…”

“…Inhibition of PTN expression in mouse embryonic pancreatic primordia explants prevented full maturation of endocrine precursors with impaired insulin and glucagon expression [ 727 ]. Further studies suggest a role for PTN in β-cell proliferation and regulation of glucose homeostasis [ 728 , 729 ]. BDNF has been shown to bind to the TrkB.T1 receptor on β-cells, resulting in increased GSIS [ 247 ].…”
