Plasmid transfer of broad-host-range plasmid RP1 from marine Vibrio sp. strain S14 to marine strain SW5 under optimum conditions on the surface of nutrient plates was improved 2 orders of magnitude by using the plasmid transfer process to select an SW5 recipient more efficient than the wild type in receiving and/or maintaining the plasmid. This recipient strain, SW5H, was used to form biofilms under flow conditions on the surfaces of glass beads in reactors. The S142(RP1) donor strain was introduced to the reactors after either 48 or 170 h of biofilm formation, and production of transconjugants in the aqueous phases and biofilms without selection pressure was assessed. Plasmid transfer to the recipient cells in the biofilm was detected for biofilms formed for 170 h but not in those formed for 48 h. The plasmid transfer frequency was significantly higher ( P < 0.05) among cells attached to the bead surfaces in the biofilm than among cells in the aqueous phase.

We have used triparental matings to demonstrate transfer (mobilization) of the nonconjugative genetically engineered plasmid pHSV106, which contains the thymidine kinase gene of herpes simplex virus cloned into pBR322, from Escherichia coli HB101 to an environmental isolate of Enterobacter cloacae in sterile drinking water. This is the first demonstration of a two-step mobilization of a genetically engineered plasmid in any type of fresh water, including drinking water. Transfer was mediated by R plasmid R100-1 of E. coli ED2149(R100-1). Matings in drinking water at 15, 25, and 35°C yielded recombinants, the number of which increased with increasing temperature. Numbers of recombinants obtained were 2 orders of magnitude lower than those obtained from matings in Trypticase soy broth. High concentrations of parental organisms (2.6 x 108 to 2.0 x 109 CFU/ml) were required. During 1 week of incubation in drinking water, numbers of parental organisms and recombinants resulting from mobilization remained constant in the absence of indigenous organisms and declined in their presence. Using oligonucleotide probes for the cloned foreign DNA (thymidine kinase gene) and plasmid vector DNA (ampicillin resistance gene), we demonstrated that both genes were transferred to E. cloacae in the mobilization process. In one recombinant selected for detailed study, the plasmids containing these genes differed in size from all forms of pHSV106 present in E. coli HB101(pHSV106), indicating that DNA rearrangement had occurred. This recombinant maintained its plasmids in unchanged form for 15 days in drinking water. A second rearrangement occurred during serial passage of this recombinant on selective media. The possibility of rearrangments complicates risk assessment because antibiotic resistance gene DNA and cloned DNA can become dissociated, making the foreign DNA more difficult to detect.

The environmental plasmid pQKH6 was transferred conjugatively between strains of Pseudomonas putida at mean frequencies of up to 8.4x10(-4) within pilot-scale sewage filter beds. This frequency was 10-fold higher than that reported previously for this environment and was probably due to seasonal temperature changes. Many (45%) of the plasmids isolated subsequently from the filter beds had restriction fragment length polymorphism (RFLP) profiles that differed from that expected for pQKH6. RFLP analysis revealed structural rearrangements occurring within a particularly restriction-site-rich region of the plasmid. Although no evidence was obtained showing the indigenous invertebrate populations within the filter beds to influence the rate of gene transfer, pQKH6 was transferred with frequencies of up to 1.6x10(-2) within the guts of the filter-bed-dwelling Sylvicola fenestralis larvae during laboratory experiments. This transfer was strongly influenced by donor to recipient ratios. Laboratory experiments also showed that Serratia fonticola survived better within invertebrate guts than P. putida. This evidence, along with experiments showing that S. fonticola could participate in pQKH6 transfer within filter-bed biofilm, identify this bacterium as a better model than P. putida for examining the effect of invertebrates on gene transfer.
