In vitro direct plant regeneration of lucerne was achieved by simultaneous application of thidiazuron (TDZ) and 6-benzyladenine (BA) in Murashige and Skoog (MS) medium. Seedlings were germinated and grown for 6 d on growth regulator-containing MS medium. The shoot tip, consisting of the apical meristem along with parts of the cotyledonary leaves and hypocotyl, was then cultured on a medium containing the growth regulator(s). Adventitious budding of the shoot tip was promoted synergistically by treatment with TDZ and BA, and a maximum of thirty-five shoots per explant was obtained on a medium supplemented with 2 mg L À1 TDZ and 1 mg L À1 BA. Plant regeneration frequency varied from 67 to 93%, and five Indian lucerne cultivars responded well to the regeneration protocol. The Agrobacterium-mediated transformation frequency from co-cultivated explants was 13% following multiple shoot induction. Southern analysis of the T 0 plants and T 1 progenies confirmed stable inheritance of the hpt marker gene. Agrobacterium infection of the explant caused a significant reduction in the plant regeneration frequency (23%) and the number of shoots induced (11) when compared with uninfected explants. A single shoot tip provided sufficient material to regenerate and establish twenty-seven lucerne plants, whereas only nine plants could be regenerated from an Agrobacterium co-cultivated explant. This transformation protocol could represent a valuable improvement over existing ones for lucerne.

Key message Tissue cultures are the only effective way to proliferate this valuable species (Daphne mezereum). These researches may help in solving the problem of reproduction of other woody plants. Abstract Daphne mezereum L. commonly known as February daphne or mezereon is a decorative shrub from the family Thymelaeaceae. It is a valuable nursery plant due to its high ornamental values, but its propagation by traditional methods is ineffective, therefore, little profitable. Micropropagation may be a good alternative way to produce this ornamental shrub. The experiment aimed to examine the effect of cytokinins (meta-Topolin, benzyladenine, and zeatin) and other growth regulators (gibberellic acid GA 3 and auxin 1-naphthalene acetic acid NAA) added to the Murashige and Skoog (MS) and Woody Plant Medium (WPM) nutrients on regeneration of explants of D. mezereum 'Alba'. In the all treatments containing plant growth regulators (PGR), 100% regeneration of explants has been observed. The highest number of shoots, after 6 weeks of culture, was produced on MS medium with 1 mg•L −1 BA and 0.1 mg•L −1 NAA. The longest shoots were produced on MS medium with 0.1 mg•L −1 of gibberellic acid (GA 3) and 1 mg•L −1 of meta-Topolin (mT). Generally, the type and concentration of plant growth regulators had an essential effect on regeneration and growth of shoots of Daphne mezereum 'Alba' in the in vitro culture.

Background: The study showed that phytochrome A (phyA) is involved in alfalfa fall dormancy regulation. To provide the scientific basis for using fall dormancy to predict new varieties’ dormancy class from the molecular level, cultivate alfalfa varieties with cold solid resistance and weak fall dormancy and improve the fall dormancy level of existing excellent types, we could study its function through gene silencing. But the genetic transformation rate is not high, which affects the research progress. Methods: To optimize the genetic transformation system in alfalfa (Medicago sativa L.), a binary plasmid vector pART27-phyA was transferred into alfalfa in Agrobacterium tumefaciens-mediated way. To improve transformation efficiency, we optimized the mediums by screening different Kan (Kanamycin) concentrations, NAA (a-Naphthylacetic acid) concentrations and adding YE (yeast extracts). Result: The results showed that using cotyledon nodes as explants, the proper concentration of Kan antibiotics resistance screening was 100 mg/L, the appropriate concentration of NAA for inducing cotyledon node differentiation was 0.1 mg/L and 0.2 g/L YE was suitable to add on the root induction medium; The expression of phyA in terminal buds and leaves of transformed plants of T1 and T2 generations of phyA in Reindeer was significantly reduced (P less than 0.05), but the expression trend in roots and stems had decreasing trend with no significant changes.
