Persistent hyperglycemia and elevated levels of free fatty acids (FFA) contribute to oxidative stress, a proximate cause for the onset and progression of diabetes and its complications. The present study was hypothesized to evaluate the anti-diabetic potential of Rosmarinic acid (RA) during high-fat diet (HFD)-streptozotocin (STZ)-induced type 2 Diabetes (T2D) in wistar albino rats. Oral administration of RA (100 mg/kg b.w) significantly (p < 0.05) increased the insulin sensitivity index (ISI0,120), while the levels of blood glucose, HbA1c, advanced glycation end products (AGE), TNF-α, IL-1β, IL 6, NO, p-JNK, P38 MAPK and NF-κB were significantly reduced, with a concomitant elevation in the plasma insulin levels in diabetic rats. Furthermore, RA treatment significantly (p < 0.05) reduced the levels of triglycerides, FFA and cholesterol in serum, and reduced the levels of lipid peroxides, AOPP's and protein carbonyls in the plasma and pancreas of diabetic rats. The diminished activities of pancreatic superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione-S-transferase (GST) and the decreased levels of plasma ceruloplasmin, vitamin C, vitamin E and reduced glutathione (GSH) in diabetic rats were also significantly (p < 0.05) recovered upon RA treatment denoting its antioxidant potential which was confirmed by Nrf-2, hemeoxyenase (HO-1) levels. Histological, ultrastructural and immunohistochemical data demonstrate that oral administration of RA protects pancreatic β-cells from oxidative niche in HFD-STZ-induced experimental diabetes. Our findings suggest that the oral treatment with RA alleviates pancreatic β-cell dysfunction and glucolipotoxicity-mediated oxidative stress during HFD-STZ-induced T2DM, perhaps through its antioxidant potential.

Background: Glycation induced cross-linking of proteins are associated with chronic diabetic complications. Inhibition of protein glycation is one of the therapeutic approaches to prevent the progression of diabetic complications. Objective: Objective of this study was to establish a simple method to identify medicinal plants which can inhibit glycation induced protein cross-linking. Methods: Lysozyme was incubated at 37°C up to 4 weeks with different concentrations of glucose, fructose and ribose in sodium phosphate buffer (pH 7.4). Appropriate controls and blanks were carried out. Aminoguanidine (AG) was used as the standard inhibitor. Water extracts of Bryophyllum pinnatum leaves, Coriandrum sativum seed and Murraya koenigi leaves were used as potential inhibitors. Aliquots were removed from the incubation mixtures at intervals and analyzed for the presence of AGE induced protein cross-links, using SDS-PAGE. Appearance and the intensity of high molecular weight products were compared. Results: Extent of cross-linking was dependent on the sugar concentration. Cross-linking was slowest in the presence of glucose and fastest in the presence of ribose. AG inhibited glycation induced protein cross-linking in the presence of all three sugars. B. pinnatum leaves, C. sativum seeds and M. koenigi leaves inhibited protein cross-linking in the presence of sugar. This inhibition was greater than that of AG. Conclusions: We have established a simple SDS-PAGE method to identify medicinal plants which inhibit glycation induced protein cross-linking. We also demonstrated the effectiveness of B. pinnatum leaves, C. sativum seed and M. koenigi leaves in inhibiting glycation induced protein cross-linking in vitro. Background: Protein glycation is a major cause of chronic diabetic complications. Medicinal plants with protein glycation inhibitory potential will be benefi cial to prevent or delay diabetic complications. Objective: The objective of the study was to analyze protein glycation inhibitory potential of ten plants among which nine are well known for their antidiabetic effects.Methods: Methanol extracts were prepared using parts from nine plants namely, Coccinia grandis, Ficus racemosa, Gymnema lactiferum, Gymnema sylvestre, Musa paradisiaca, Phyllanthus debilis, Phyllanthus emblica, Strychnos potatorum and Tinospora cordifolia. Dried latex of Pterocarpus marsupium was used without further extraction. Glycation inhibitory potential of the extracts was analyzed using bovine serum albumin (BSA) and fructose, incubated in the presence or absence of plant extracts for 4 week. Appropriate controls and blanks and the standard inhibitor aminoguanidine were included. A novel native polyacrylamide gel electrophoresis (PAGE) method established recently was used to detect the effect of plant extracts on the BSA glycation. Results: Previously we have demonstrated that the BSA migration towards the anode is increased proportionate to the degree of glycation. This increase was comparatively less when the glycation was inhibited. Accordingly, most promising antiglycation effects were observed with P. debilis (whole plant), P. emblica (fruit) and P. marsupium (latex). F. racemosa (stem bark) showed the highest inhibition among the remaining extracts. G. sylvestre (leaf), M. paradisiaca (yam) and T. cordifolia (leaf) also showed promising inhibition. C. grandis (leaf) and S. potatorum (seed) showed lower inhibition. Lowest inhibition was observed with G. lactiferum (leaf).Conclusion: We have demonstrated the antiglycation potential of some antidiabetic plants, using a novel method developed by us recently.
