Abstract: Partial inhibition of PI3K is one of the best-validated and evolutionary conserved manipulations to extend longevity. The best known health beneficial effects of reduced PI3K are related to metabolism and include increased energy expenditure, reduced nutrient storage, and protection from obesity. We have previously shown that a dual chemical inhibitor of the alpha and delta PI3K isoforms (CNIO-PI3Ki) reduces obesity in mice and monkeys, without evident toxic effects after long-term treatment. Here, we dissect … Show more

“…As discussed in our respective article, there are numerous reports in the literature demonstrating a profound lack of tissue specificity for the aP2-Cre strain, which complicates interpretation of phenotypes. Importantly, the phenotypes described in our previous and present work are consistent with those reported for long term pharmacological inhibition of p110α 20,21 , which adds additional weight to the validity of our findings.…”

“…Furthermore, adipose tissue-specific inactivation of p110α resulted in leaner mice due to increased energy expenditure 19 . Importantly, long-term pharmacological inhibition of p110α has previously been shown to protect mice and rhesus monkeys from diet-induced obesity, highlighting both the utility of PI3K inhibitors in metabolic studies and their therapeutic potential 20,21 . Although a worm ortholog of class I PI3K was the first gene ever reported to affect the rate of ageing 22 , the specific roles of PI3K in the context of processes associated with ageing have barely been investigated.…”

“…Often components of this and related phospholipid signaling pathways are tumor suppressors [ 124 , 125 ]. Multiple components of the PI3K/PTEN/AKT/mTORC1 pathway have and are being considered for potential targeting in human cancer, e.g., p70S6K in breast cancer [ 126 ], as well as, obesity [ 127 ].…”

“…Obesity is the major risk factor that drives type 2 diabetes epidemics by promoting insulin resistance and loss of pancreatic b-cell mass and function. Preclinical studies show that the inhibition of class 1 PI3K reduces adiposity and improves blood glucose homeostasis in rodents and primates (1)(2)(3)(4)(5)(6)(7)(8). Thus, pharmacologic inhibition of specific class 1 PI3Ks may be an effective treatment for obesitydriven diabetes in humans.…”
