The paper presents an investigation into the effects of endurance training and testosterone enanthate (TE) treatment on rat blood serum by fluorescence spectroscopy. Blood serum was collected from adolescent male Wistar rats randomized between the untrained controls and trained animals, either steroid untreated or receiving I.M. injections (8 or 80 mg/kgBW) of testosterone enanthate. Fluorescence measurements were performed for undiluted or 20-and 500-fold diluted serum and analysed separately in the ultraviolet and visible light regions. In both regions the impact of the TE administration was more pronounced than the effect of endurance training. Moreover, in the visible region of spectrum, originated from a number of fluorescent metabolites, the influence of studied factors was more noticeable. The opposite effect of endurance training and administration of TE on serum emission was found for a lower (8 mg/kgBW) dose of testosterone, whereas a high-dose TE (80 mg/kgBW) treatment resulted in a return of fluorescence intensity.

Differential scanning calorimetry (DSC) was applied to evaluate whether endurance training and/or testosterone treatment affect thermal behavior of rat serum. Thirty-six adolescent male Wistar rats were divided into six groups: three untrained and three trained groups, receiving weekly 0, 8, or 80 mg kg BM -1 of testosterone enanthate (TE) each. Total testosterone was significantly higher in both groups treated with the higher TE dose. The rats' body mass and the levels of serum glucose, uric acid, and free fatty acids were additionally determined. The effect of TE treatment was clearly visible in DSC curves for sedentary rats and much less for trained rats. The differences between averaged endothermic denaturation transitions of serum from sedentary and physically active untreated groups were within an experimental error. A statistically significant increase of the width of peak at its half height and the ratio of excess heat capacity at 75°C and in the temperature of peak maximum (C p75 /C pmax ) were observed due to TE treatment. The same two parameters were significantly lower for physically active rats. Significantly lower values of the enthalpy of serum denaturation (DH) and the first moment of the transition with respect to the temperature axis (M 1 ) were observed for endurance-trained than for sedentary rats.

Hormonal changes due to menopause can cause various health problems including weight gain and depressive symptoms. Multiple lines of evidence indicate that oestrogen receptors (ERs) play a major role in postmenopausal obesity and depression. However, little is known regarding the ER subtype-specific effects on obesity and depressive symptoms. To delineate potential effects of ERβ activation in postmenopausal women, we investigated the effects of a novel oestrogen receptor β-selective ligand (C-1) in ovariectomized mice. Uterine weight, depressive behaviour, and weight gain were examined in sham-operated control mice and ovariectomized mice administered placebo, C-1, or 17β-oestradiol (E2). Administration of C-1 or E2 reduced body weight gain and depressive-like behaviour in ovariectomized mice, as assessed by the forced swim test. In addition, administration of E2 to ovariectomized mice increased uterine weight, but administration of C-1 did not result in a significant increase in uterine weight. These results suggest that the selective activation of ERβ in ovariectomized mice may have protective effects against obesity and depressive-like behaviour without causing an increase in uterine weight. The present findings raise the possibility of the application of ERβ-ligands such as C-1 as a novel treatment for obesity and depression in postmenopausal women.
