Phosphorus Dendrimers as Nanotools against Cancers

Abstract: This review concerns the use of dendrimers, especially of phosphorhydrazone dendrimers, against cancers. After the introduction, the review is organized in three main topics, depending on the role played by the phosphorus dendrimers against cancers: (i) as drugs by themselves; (ii) as carriers of drugs; and (iii) as indirect inducer of cancerous cell death. In the first part, two main types of phosphorus dendrimers are considered: those functionalized on the surface by diverse organic derivatives, including kn… Show more

“…For the synthesis of dendritic polymers, anionic [ 31 , 32 , 33 , 34 ], click-chemistry [ 35 ] and organometal-catalyzed [ 36 ] techniques have already been employed and are reported in the literature. Due to their distinctive structures, dendritic polymers and/or dendrimers exhibit unique properties, rendering them suitable to be widely used in various applications [ 37 , 38 ], such as: drug delivery [ 39 , 40 , 41 , 42 ], catalytic systems [ 43 , 44 ], membranes [ 45 ], bioimaging [ 46 , 47 ], etc.…”

“…26 To date, PPH dendrimers have been synthesized successfully with the generation up to 12, being the highest among all types of dendrimers. 27 In recent years, it has been proven to be feasible to modify different cyclic amines such as pyrrolidine, morpholine, methyl piperazine, and phenyl piperazine onto the phosphorous dendrimer surface 28 and incorporate different metal elements to form various kinds of nanoplatforms for enhanced drug 29 or gene delivery 30 toward cancer management. Several remarkable reviews by Majoral, Mignani, and Caminade et al have well described the synthesis strategy and biological applications of phosphorous dendrimers.…”

“…Several remarkable reviews by Majoral, Mignani, and Caminade et al have well described the synthesis strategy and biological applications of phosphorous dendrimers. 28,31,32 Among various modified phosphorous dendrimers that have been reported, aza-bisphosphonate (ABP)-capped phosphorous dendrimers have been a focus for their interesting self-owned bioactivities involving the immune cells. Pioneering research shows that ABP-capped dendrimers impact the state of monocytes and exert anti-inflammatory effects via inducing the expression of mannose receptor 1 and anti-inflammatory cytokines (IL-4, IL-10 and IL-13) while decreasing the expression of inflammatory markers of CD64 and CD13.…”
