Perturbations in Lineage Specification of Granulosa and Theca Cells May Alter Corpus Luteum Formation and Function

Abstract: Anovulation is a major cause of infertility, and it is the major leading reproductive disorder in mammalian females. Without ovulation, an oocyte is not released from the ovarian follicle to be fertilized and a corpus luteum is not formed. The corpus luteum formed from the luteinized somatic follicular cells following ovulation, vasculature cells, and immune cells is critical for progesterone production and maintenance of pregnancy. Follicular theca cells differentiate into small luteal cells (SLCs) that produ… Show more

“…At E18.5, 72% of cells stained positive for GATA4/FOXL2, with 1.7% of cells stained with the oocyte marker DDX4 from counts of 200 cells repeated across n = 3 biological replicates. Dissociated cells from this timepoint showed no PTCH1 expression, consistent with a lack of theca cells present in the embryonic mouse ovary ( Abedel-Majed et al , 2019 ). From cells dissociated from the 12 weeks ovary timepoint, 64% of cells stained positive for GATA4/FOXL2, with 1.6% of cells stained with the oocyte marker DDX4.…”

“…The effects of BCR on ovarian functions were evaluated in the following aspects. It is known that the estrous cycle and ovarian corpus lutea from the last ovulation can reflect the status of ovulation of rats (Abedel-Majed et al, 2019;Liman and Ates, 2020). The growing follicle count, and especially the antral follicle count, is a reliable method for predicting the total number of mature follicles and pregnancy rate (Keane et al, 2017).…”

“…The methylation level of genes at secondary follicles was set as 100 %. The methylation status did not change dramatically in the genes, Ccnd2, Foxo3, Fshr, Ar, that are highly expressed in secondary follicles and maintained in preovulatory follicles [3,27]. However, the methylation levels did dramatically decrease to about half in the genes, Amhr2, Gja1, Nr5a1, Lhcgr in which the transcription is known to be increased after eCG injection [3,28,29].…”
