Peroxisome proliferator-activated receptor-γ–coactivator 1α (PGC-1α) gene expression in chronic kidney disease patients on hemodialysis: relation to hemodialysis-related cardiovascular morbidity and mortality

Abstract: Our study suggests that ESRD/HD patients might have oxidative mitochondrial dysfunction, which may be partially responsible for CKD-related cardiovascular complications. Pharmacological modulation of PGC-1α might be a promising therapeutic tool to reduce oxidative stress-related complications in ESRD/HD patients.

“…Oxidative stress may be considered as a means for predicting morbidity and mortality in CKD patients. 10,11 CKD patients have low levels of antioxidants and high concentrations of pro-oxidants. 12,13 Meanwhile, oxidative stress and inflammation are interwoven; it is not only oxidative stress that acts as a trigger for inflammatory response, but also inflammation in turn recruits phagocytes into an injured region, leading to production of oxygen radicals.…”

“…Considerable evidence has indicated that hemodialysis patients are often accompanied by lipid metabolism disorders, cardiovascular complications in patients with one of the important factors [1-5]. However, the etiology and underlying mechanism are not entirely clear.…”

“…Peroxisome proliferator-activated receptor γ (PPARγ) coactivator 1 (PGC-1α), the energetic regulator, is acknowledged as a critical signaling pathway regulating COX6c expression [ 25 ]. As a powerful regulator of cellular energy metabolism, PGC-1α is primarily expressed in tissues and organs rich in mitochondria, such as the heart, liver, kidney, brain, brown adipose tissue, and skeletal muscle.…”
