Abstract: Recycling of carbon by the photorespiratory pathway involves enzymatic steps in the chloroplast, mitochondria, and peroxisomes. Most of these reactions are essential for plants growing under ambient CO(2) concentrations. However, some disruptions of photorespiratory metabolism cause subtle phenotypes in plants grown in air. For example, Arabidopsis thaliana lacking both of the peroxisomal malate dehydrogenase genes (pmdh1pmdh2) or hydroxypyruvate reductase (hpr1) are viable in air and have rates of photosynthe… Show more

“…Using a candidate gene approach, the gene encoding phosphoglycolate phosphatase was revealed based on the characteristic photorespiratory phenotype of the knockout mutant (i.e., nonviability in normal air but normal growth under elevated CO 2 concentrations) (Schwarte and Bauwe, 2007). Contrary to the other core photorespiratory enzymes, deletion of peroxisomal HPR1 does not lead to ambient air sensitivity but does increase the stoichiometry of photorespiratory CO 2 release (Cousins et al, 2011). Identification of a second HPR (HPR2) suggests the existence of an efficient NADPH-dependent cytosolic bypass (Timm et al, 2008).…”

“…Artificial constraints on respiratory and photorespiratory metabolism can have distinct effects on photosynthetic parameters (Nunes-Nesi et al, 2005Cousins et al, 2011;Timm et al, 2011Timm et al, , 2012aEisenhut et al, 2013). To find out whether such effects were also apparent in the mtLPD overexpressor plants, we examined the photosynthetic-(photo)respiratory gas exchange in more detail.…”

“…However, at 25 °C the post-illumination burst (PIB) and 12 CO 2 release following a saturating 13 CO 2 injection both scale with photorespiratory CO 2 release, providing an estimate of the CO 2 component of α (Doehlert et al, 1979; Delfine et al, 1999; Loreto et al, 2001; Cousins et al, 2008, 2011). Additionally, v o can be estimated using isotopic exchange of 18 O 2 and 16 O 2 , but these measurements are subject to the assumptions of O 2 exchange outlined previously and discussed in the theory section below (Canvin et al, 1980; Badger, 1985; Cousins et al, 2008, 2011). Recently, measurements of 12 CO 2 release and 18 O 2 and 16 O 2 exchange indicated an increase in α in Arabidopsis thaliana lacking both isoforms of peroxisomal malate dehydrogenase ( pmdh1pmdh2 ) and peroxisomal hydroxypyruvate reductase ( hpr ) (Cousins et al, 2008, 2011).…”
