This research was conducted to investigate the effect of various concentrations of lactose supplementation in Tris extender for maintaining the quality of Etawa crossbreed goat epididymal spermatozoa stored at 3-5° C. Semen in the control group was diluted with a tris extender containing 20% egg yolk without lactose. Semen in the test groups was diluted with a tris extender containing 20% egg yolk and added with 0.3% (0.3 g per 100 mL extender) and 0.6% lactose for group TL1 and TL2, respectively. Parameters evaluated of the fresh epididymal spermatozoa were motility, concentration, percentage of live, and abnormality of spermatozoa, while for diluted-spermatozoa were motility and percentage of live spermatozoa. Spermatozoa observation was conducted until it reaches 40% motility. The results showed that the mean percentage of motility, live sperm, concentration, and abnormality of epididymal spermatozoa were 70%; 81%; 3,220x106 cells/mL; and 4.30%, respectively in all group. After dilution, the percentage of motility and live spermatozoa were also 70% and 81.00±1.58%, respectively in all groups. The decreasing of spermatozoa motility was observed on day 4 of storage, in which percentage of spermatozoa motility in control group (40.00±0.00%) was significantly lower (P<0.05) than those in TL1 (44.00±2.24%) and TL2(45.00±0.00%) groups. Percentage of live spermatozoa in control (63.20±2.68%) was not significantly different (P>0.05) than TL1(65.40±1.95%) and TL2 (65.60±1.95%). In conclusion, the supplementation of lactose into Tris extender could maintain the epididymal spermatozoa of Etawa crossbreed for 3 days of storage at 3-5° C.

ABSTRAKTujuan penelitian ini adalah untuk mengetahui pengaruh pencucian semen dan kadar lesitin kacang ⁰ kedelai terhadap motilitas dan viabilitas sperma domba yang disimpan pada suhu 5 C. Semen tampung dari tiga ekor domba jantan dewasa dengan vagina buatan. Semen dievaluasi secara makroskopis dan mikroskopis. Semen dibagi dalam sepuluh tabung, semen dari lima tabung pertama tidak dicuci (W0) dan diencerkan dengan extender 0%, 1%, 2%, 3% dan 4% lesitin kacang kedelai (L0, L1, L2, L3 dan L4). Semen dari lima tabung lainnya dicuci (W1) kemudian masing-masing diencerkan dengan kadar ⁰ extender yang sama. Sampel semen yang telah diencerkan disimpan pada suhu 5 C, kemudian motilitas dan viabilitas sperma dievaluasi setiap hari. Interaksi dari pencucian semen dan kadar lesitin kacang kedelai tidak berbeda nyata (P>0.05) tetapi secara tunggal faktor kadar lesitin kacang kedelai berpengaruh nyata (P<0.05) dan faktor pencucian semen tidak berpengaruh nyata (P>0.05) terhadap motilitas progresif dan viabilitas sperma domba. Ekstender terbaik adalah 3% lesitin kacang kedelai (L3) dengan persentase motilitas progresif dan viabilitas spermatozoa masing-masing 63.18 ± 3.61% dan 71.76±2.32%. Semen was collected from three mature rams by artificial vagina. Semen was evaluated by macroscopic and microscopic. Semen was divided into ten tubes, the semen of first five tubes was unwashed (W0) and diluted with extender 0%, 1%, 2%, 3% and 4% soybean lecithin (L0, L1, L2, L3 and L4). Semen of another five tubes were washed (W1) then each diluted with similar extender levels. The diluted semen samples were stored at ⁰ 5 C and the sperm motility and viability were evaluated each day. The interaction of the semen washing and soybean lecithin levels were no significant differences (P>0.05) but the single factor of soybean lecithin level was significant differeNces (P<0.05) and the single factor of semen washing were no significant differences (P>0.05) on progressive motility and viability of ram sperm.The best extender was 3% soybean lecithin (L3) with the percentage of ram sperm progressive motility was 63.18 ± 3.61% and viability was 71.76±2.32%.

Rizal (2006) Namun apabila dibandingkan antara pengencer air kelapa tuavaietas merah (P1) dengan kelapa tuavarietashijau (P2) menunjukkan bahwa air kelapa hijau lebih mampu mempertahankan motilitas individu spermatozoa sampai hari ke-3 dengan nilai rataan secara berurutan P0(65±7,8); P2(33,5±24,4) dan P1 (19±28,4). Artificial Insemination using liquid semen is technology reproduction can improve goat productivity without liquid nitrogen. The Purpose of this research was to determine differences of old coconut water varieties of red (Cocos rubescens) and green (Cocos viridis) as diluent Boer goat semen, based on motility, viability and spermabnormalities during storage at 3-50C. The materials used for this research was Boer goat fresh semen as much as 3 heads were collected 2 times a week. Experimental Design. The treatment were devided into three Treatmen, there were: P0 (Tris+ 20% egg yolk); P1 (old coconut water varieties red + 20% egg yolk); P2 (old coconut water varieties green + 20% egg yolk). The data were analyzed by randomized block design, if the result are different or significantly different then continued to be tested with Duncan's multiple range test. The result showed that during storage until days 4, P0 has the best result on motility (40,5 + 1,1 %), viability (45,18+ 1,56 %), and abnormality (1,89+ 0,59 %). While on the diluent using old coconut water green variety better then red variety. On day of 2 P1 and P2 have motility (61 + 1,3%) and (51+ 7,5%); Viability (67,14+17,99%) and (55,37+10,66%), abnormality (1,23 + 1,02%) dan (1,70+ 0,99%).
