Pemphigus vulgaris. Superior sensitivity of monkey esophagus in the determination of pemphigus antibody

Abstract: This study demonstrates that monkey esophagus is a more sensitive substrate than guinea pig esophagus for the demonstration of serum antibodies, in patients with pemphigus vulgaris. In this study, for example, five patients with active pemphigus who had demonstrable pemphigus antibodies on monkey esophagus substrate failed to demonstrate pemphigus antibody activity when guinea pig esophagus was employed as a substrate. Furthermore, pemphigus antibody titers determined on monkey esophagus substrate frequently e… Show more

“…Total IgG concentration was measured by nephelometry using monospecific goat anti-human IgG (Beckman Instruments, Missasauga, Ontario, Canada). Pemphigus Ab titers were measured by indirect immunofluorescence using monkey esophagus epithelium as the tissue substrate (22). As a negative control, IgG fractions were isolated and removed from PV plasma using protein A (PA).…”

“…Sera were collected 24 h after PV IgG treatment, PA treatment (IgG depleted), or sham treatment. Indirect immunofluorescence studies were performed according to standard techniques (22,26). Cryostat sections (5 m) of monkey esophagus were employed as substrate, washed for 10 min in PBS, incubated for 30 min with different concentrations of sera (1:1-1:600), washed in PBS for 15 min, labeled with FITC-conjugated F(abЈ) 2 fragment of rabbit anti human IgG (Dako) for 30 min, and then washed again in PBS for 15 min.…”

“…Serum from one PV patient was then diluted in PBS over a range of eight doubling dilutions from 1:10 to 1:1280 [11]. The final titer that was obtained from ME substrate was used as a reference titer for this serum.…”

“…For IIF technique, patient serum is used to identify circulating IgG antibodies to the intercellular spaces of keratinocytes of substrate. Monkey esophagus (ME) and guinea pig esophagus are well-accepted substrates for pemphigus vulgaris (PV) and pemphigus foliaceous (PF), respectively [11,14]. Sensitivity of IIF on ME ranged from 81 to 100 % [2,14,[32][33][34]37].…”
