Abstract: Although platelet-derived growth factors (PDGFs) and receptors (PDGFRs) are abundantly expressed in the central nervous system, their functions largely remain elusive. We investigated the role of PDGFR-β in tissue responses and functional recovery after photothrombolic middle cerebral artery occlusion (MCAO). In the normal adult mouse brain, PDGFR-β was mainly localized in neurons and in pericyte/vascular smooth muscle cells (PC/vSMCs). From 3 to 28 days after MCAO, postnatally induced systemic PDGFR-β knockou… Show more

“…28 While astroglia can provide a favorable environment for regeneration, 29 fibroblastlike cells are known to segregate astrocytes from the lesion via ephrin-B2/EphB2 signaling. 30 Conditional deletion of PDGFRb during stroke resulted in larger infarcts and disruption of astroglial scar formation, 31 suggesting a potential crosstalk between PDGFRb þ cells and astroglia. Inflammatory cells such as macrophages, neutrophils, and lymphocytes modulate fibrosis via the release of interleukins and growth factors, including PDGF and transforming growth factor-b.…”

“…21,22 Furthermore, it is experimentally suggested that efficient fibrotic formation within areas of infarct may promote peri-infarct astrogliosis. 23 Thus, if these repair processes are associated with functional recovery, apparent infarct volumes at early phases may not be suitable for predicting functional outcome. To date, few papers have focused on the effects of reperfusion on poststroke repair processes in both animals and human. In the present study, we hypothesized that early reperfusion might elicit efficient tissue repair, such as intrainfarct fibrosis and peri-infarct astrogliosis, and lead to functional recovery even if it does not prevent neuronal death within the ischemic areas.…”

“…При ишемическом инсульте в эксперименте введение PDGF оказывает нейропротективное действие, пре-пятствует развитию отека ткани головного мозга, усиливает ангиогенез как в зоне повреждения, так и на границах со здоровой тканью [137,138].…”
