ABSTRACT:The antibacterial activity present in the s k~n mucus of turbot Scophthalmus maximus, seabream Sparus aurdtd and seabass Dicentrarchus labrdx against Pasteul-ella piscicida and Flex~bac-ter maritimus was evaluated. Using assays on agar plates, none of the mucus samples from the above fish showed any antibacterial activity against E maritimus isolates. Turbot mucus inhibited the growth of the P piscicida but mucus from seabream and seabass did not. Assays in liquid systems to determine the survival of the above pathogens In the presence of skin mucus corroborated the results obtained by the agar plate method. The bactericidal properties of the mucus were lost after heat treatment at pH 3.5 and all skin mucus samples displayed act~vity against Staphylococcus aureus ATCC 25923, a straln resistant to lysozyme. These findings indicated that thermolabile substances other than lysozyme were responsible for the antibacterial activity in mucus of marine fish. Enzymatic and heat treatments of the mucus also showed that factors other than complement were involved and that the active component(s) was likely a glycoprotein. Regardless of the source of isolation and degree of virulence, all P piscicida and E maritimus strains adhered strongly to the skin mucus of the 3 fish species tested. Taking all of the foregoing results into consideration, it appears that whereas a possible portal of entry for E maritlmus Into the fish body is the skin, in P piscicida another pathway must be involved.

The increasing problem of antibiotic resistance in common pathogenic bacteria and the concern about the spreading of antibiotics in the environment bring the need to find new methods to control fish pathogens. Phage therapy represents a potential alternative to antibiotics, but its use in aquaculture requires a detailed understanding of bacterial communities, namely of fish pathogenic bacteria. Therefore, in this study the seasonal dynamics of the overall bacterial communities, microbiological water quality and disease-causing bacteria were followed in a marine aquaculture system of Ria de Aveiro (Portugal). Analysis of the bacterial diversity of the water samples by denaturing gradient gel electrophoresis (DGGE) of 16S rRNA gene fragments indicates that the bacterial community structure varied seasonally, showing a higher complexity during the warm season. The diversity of the main fish pathogenic bacteria, assessed by DGGE targeting the Vibrio genus, showed lower seasonal variation, with new dominating populations appearing mainly in the spring. Bacterial indicators, faecal coliforms and enterococci, enumerated by the filter-membrane method, also varied seasonally. The fluorescent in situ hybridization (FISH) results showed that the specific groups of bacteria varied during the study period and that the non-indigenous Enterobactereaceae family was the most abundant group followed by Vibrio and Aeromonas. The seasonal variation detected in terms of density and structure of total and pathogenic bacterial communities demonstrates the need for a careful monitoring of water through the year in order to select the suitable phages to inactivate fish pathogenic bacteria. The spring season seems to be the critical time period when phage therapy should be applied.

Serological differences of Photobacterium damsela subsp. piscicida (= Pasteurella piscicida) isolates were determined using yellowtail (Seriola quinqueradiata) and rabbit antisera. In Oita, Japan, from June to October, 1995, Ph. damsela piscicida was isolated from net-pen cultured diseased yellowtail and sera were collected from fish cultured in the same net-pen.Antisera against formalin-killed cells of three representative isolates were raised in yellowtail and rabbits.The yellowtail antisera were absorbed with homologous or heterologous isolates and used for the agglutinating antibody absorption test. Crude lipopolysaccharides were extracted from the isolates and analyzed by immunodiffusion with rabbit antisera. As a result, three types of antigenic differences were revealed among the isolates by the agglutinating tests with the sera of naturally infected fish and by the absorption tests using yellowtail antisera. Some common and distinctive antigens of the isolates were found by immunodiffusion analysis with rabbit antisera.
